Are genetic variations in IL‐21–IL‐23R–IL‐17A cytokine axis involved in a pathogenic pathway of rheumatoid arthritis? Bayesian hierarchical meta‐analysis

Abstract: Inflammatory cytokines have been established to be involved in the pathogenesis of rheumatoid arthritis (RA). The genetic polymorphisms in the interleukin (IL) 23 receptor (IL23R), IL21, and IL17 have been associated with RA risk. However, there is no conclusive understanding of the genes encoding the immunoinflammatory IL‐21–IL‐23R–IL‐17A pathway in RA aetiopathogenesis. This meta‐analysis was conducted to attain this goal. A comprehensive literature search was conducted in Scopus and PubMed to look for the r… Show more

“…The 95% CrI is the Bayesian equivalent for standard confidence intervals. In particular, the model supposes that the mean of the Log ORs has a low-informative normal distribution (mean = 0, variance = 100) and the variance of the Log ORs has a low-informative inverse-gamma distribution (0.01, 0.01) [24]. Sensitivity analyses with different choices of low-information prior distributions showed the robustness of this choice [20].…”

“…The rs12722495 polymorphism of this gene correlates with its mRNA and protein expression in stimulated monocytes, naive T-cells, and memory T-cells [ 6 ]. Additionally, according to a meta-analysis of 37 studies, a minor allele A of the rs1343151 polymorphism in the IL23R gene, which encodes the receptor for interleukin 23, is associated with a risk of developing RA [ 23 ].…”

“…Recently, Mohammadi et al also made Bayesian meta-analysis about polymorphisms of inflammatory cytokines and RA risk; however, positive results were found about genetic polymorphisms of IL23R gene, but not IL-21 and IL17A genes. Moreover, RF and ACPA status were not involved [44], which were the different and innovate about present study.…”
