Arbuscular mycorrhizal fungi play a role in protecting roots of Sophora viciifolia Hance. from Pb damage associated with increased phytochelatin synthase gene expression

Abstract: Understanding the influence of arbuscular mycorrhizal (AM) fungi on the expressions of the dominant plant-related genes under heavy metal (HM) stress is important for developing strategies to reclaim polluted sites. In this study, we cloned full-length cDNAs of phytochelatin synthase gene (PCS1) and Actin of Sophora viciifolia Hance., a predominant plant in Qiandongshan lead and zinc mine, by rapid amplification of cDNA ends. Consequently, we studied the response of SvPCS1 to Funneliformis mosseae inoculation … Show more

“…[ 37 ]. Furthermore, AMF symbiosis also induce the expression of the phytochelatin synthase gene ( PCS1 ) in a leguminous plant ( Sophora viciifolia ) that was exposed to Pb pollution [ 42 ], indicating another mechanism for Pb immobilization in the roots of plants that were enhanced by AMF.…”

“… 23 . In addition, the presence of AMF increased the expression of the phytochelatin synthase gene ( PCS1 ) in legume plant ( Sophora viciifolia ) exposed to Pb stress 24 , suggesting another mechanism of Pb immobilization in the roots of plants enhanced by mycorrhizal inoculation.…”

“…Standard and environmental DNA samples were added at 2.0 µL per reaction. The reaction was carried out on a CFX Connect Real-Time PCR Detection System (Bio-Rad, USA) using a program of 95 • C for 10 min, followed by 40 cycles of 95 • C for 30 s, 55 • C for 30 s and 72 • C for 30 s according to our previous study [42]. Specific amplification of the target gene was confirmed on a 1% (w/v) agarose gel showing the band of the expected size and melting curve analysis resulting in a single peak.…”
