Arabidopsis Voltage-Dependent Anion Channel 1 (AtVDAC1) Is Required for Female Development and Maintenance of Mitochondrial Functions Related to Energy-Transaction

Abstract: The voltage-dependent anion channels (VDACs), prominently localized in the outer mitochondrial membrane, play important roles in the metabolite exchange, energy metabolism and mitochondria-mediated apoptosis process in mammalian cells. However, relatively little is known about the functions of VDACs in plants. To further investigate the function of AtVDAC1 in Arabidopsis, we analyzed a T-DNA insertion line for the AtVDAC1 gene. The knock-out mutant atvdac1 showed reduced seed set due to a large number of undev… Show more

“…Last, O 2 uptake by the OXPHOS pathway was reduced in vdac1, and also probably in vdac3. Reduced ATP synthesis and reduced membrane potential have already been observed in vdac1 mutants [4,40], and reduced membrane potential has been found in vdac3, but also in vdac2 and vdac4 KO lines [4]. VDAC3 overexpression did not seem able to restore OXPHOS respiration in vdac1_OEV3, suggesting that VDAC1 and VDAC3 have some specialized functions that do not overlap.…”

“…The levels of VDAC1 were also higher in germinating seeds compared mature (i.e., 3-week-old) seedlings. In Arabidopsis, VDAC1 is required for maintaining the steady state of mitochondrial transmembrane potential (DC) and hence for optimal respiratory activities, and its functions were found to be critical during the reproductive stage and during early germination (Pan et al, 2014). In contrast to MatR and Porin, the steady-state levels of various other mitochondrial proteins, including Nad9, CA2, RISP, COX2, and AtpB, all increased in during plant development, with the highest accumulation levels seen in mature plants.…”

“…therein), and it is important to note that a functional interplay between these transport proteins is essential for mitochondrial activity and energy supply for the cell. Interestingly, in mammals, VDACs have been associated with PCD (Pan et al, 2014), while in plants, their contribution to cell death mechanisms remains elusive (Robert et al, 2012;Godbole et al, 2013).…”
