Arabidopsis Lectin Receptor Kinase P2K2 Is a Second Plant Receptor for Extracellular ATP and Contributes to Innate Immunity

Abstract: In animals, extracellular adenosine 5'-triphosphate (eATP) is a well-studied signaling molecule that is recognized by plasma membrane-localized P2-type purinergic receptors. However, in contrast, much less is known about purinergic signaling in plants. P2 receptors play critical roles in a variety of animal biological processes including immune system regulation. The first plant purinergic receptor, Arabidopsis P2K1 (L-type lectin receptor kinase-I.9), was shown to contribute to plant defense against bacterial… Show more

“…It is the founding member of purinoreceptor (P2K) family, with DORN1 as P2K1 (Choi et al ., 2014). Very recently, Arabidopsis lectin receptor kinase LecRK‐I.5 was shown to act as a second plant receptor for eATP and designated as P2K2 (Pham et al ., 2020). P2K2 shares 74% amino acid similarity to P2K1 and binds to ATP with similar high affinity ( Kd = 44.47 ± 15.73 n m ).…”

“…In Arabidopsis thaliana , eATP can act as a damage-associated molecular pattern (DAMP) and activates immunity signalling through the plasma membrane purinoreceptor AtDORN1 (Does Not Respond to Nucleotides1, also known as P2K1) [ 7 ]. A plasma membrane co-receptor P2K2 has recently been identified and both DORN1/P2K1 and P2K2 are lectin receptor kinases [ 8 ]. eATP-dependent but AtDORN1-independent effects have also been reported [ 9 , 10 ], pointing to the presence of other perception mechanisms.…”

“…Previous research clearly indicates that eATP is an important stress signaling compound in plants, which in Arabidopsis is recognized by the P2K1 receptor, perhaps in combination with the recently identi ed, closely related P2K2 receptor 34 . Details of importance of eATP signaling in plants are emerging, including evidence that P2K1 contributes to plant defense against pathogens 33,48,49 , ATP mediated ROS signaling, JA levels and S-acylation 33,37,49 .…”
