Arabidopsis HFR1 Is a Potential Nuclear Substrate Regulated by the Xanthomonas Type III Effector XopDXcc8004

Abstract: XopDXcc8004, a type III effector of Xanthomonas campestris pv. campestris (Xcc) 8004, is considered a shorter version of the XopD, which lacks the N-terminal domain. To understand the functions of XopDXcc8004, in planta, a transgenic approach combined with inducible promoter to analyze the effects of XopDXcc8004 in Arabidopsis was done. Here, the expression of XopDXcc8004, in Arabidopsis elicited the accumulation of host defense-response genes. These molecular changes were dependent on salicylic acid and corre… Show more

“…XopD acts as a T3S effector and has weak ubiquitin-like modifier (SUMO) isopeptidase activity (46). Overexpression of xopD from Xanthomonas in Arabidopsis leaves induces a host defense response that is dependent on salicylic acid (47). This suggests that the cysteine protease of B. elkanii USDA61 identified in the current study might be a type 3 effector and stimulate the plant immune response, eventually leading to nodule restriction in soybean varieties carrying the Rj4 allele.…”

“…Furthermore, XopD Xcc8004 binds to and deSUMOylates the basic HLH transcription factor HFR1 in subnuclear foci (Tan et al. 2015) (Fig. 7A), suggesting that XopD family members target plant transcription regulators to promote bacterial virulence.…”

“…XopD Xcc8004 , a shorter version of XopD lacking the N-terminal domain, is also a type III effector from Xanthomonas campestris pv. campestris (Xcc) acting as a SUMO protease (Tan et al, 2015). XopD Xcc8004 expression in Arabidopsis elicited host defence response genes solely dependent on its SUMO protease activity; in transgenic plants harbouring XopD Xcc8004 (C355A), no elicitation was noticed (Tan et al, 2015).…”
