Arabidopsis CHROMATIN REMODELING 19 acts as a transcriptional repressor and contributes to plant pathogen resistance

Abstract: Chromatin remodelers act in an ATP-dependent manner to modulate chromatin structure and thus genome function. Here, we report that the Arabidopsis (Arabidopsis thaliana) remodeler CHROMATIN REMODELING19 (CHR19) is enriched in gene body regions, and its depletion causes massive changes in nucleosome position and occupancy in the genome. Consistent with these changes, an in vitro assay verified that CHR19 can utilize ATP to slide nucleosomes. A variety of inducible genes, including several important genes in the… Show more

“…We next plotted the ChIP-seq signals of SYD, BRM, SPT6LΔtSH2, and SPT6L at SYD-occupied genes and also found the genome-wide concurrent signals of SYD, BRM and SPT6LΔtSH2 at TSS (Figure 1B ). To examine the specificity of co-binding between SWI2/SNF2 and SPT6LΔtSH2, we also plotted four other chromatin remodelers on the same genomic regions and found that except BAF60 (SWI/SNF family) ( 55 ), other three chromatin remodelers PHOTOPERIOD INDEPENDENT EARLY FLOWERING 1 (PIE1, SWI2/SNF2-related 1 family) ( 56 ), CHROMATIN REMODELING 11 (CHR11, ISWI family) ( 56 ), and a putative SNF2-related chromatin remodeler CHR19 ( 57 ) were weakly or even not enriched at TSS (Figure 1B ). This result suggests that SPT6LΔtSH2 may prefer to interplay with SWI/SNF complex at TSS.…”

“…Previous studies have shown that OsCHR725 ( OsBRHIS1 ) plays a critical role in the SA-independent disease resistance ( Li et al, 2015 ), suggesting that the cis -acting elements in the promoter are coordinated with the gene expression and function. AtCHR19 acts as a transcriptional repressor and contributes to plant pathogen resistance ( Kang et al, 2022 ). OsCHR714 , the homologous gene of AtCHR19 , was significantly upregulated at the 12 hpi of M. oryzae and all treatment timelines of salt and drought stress.…”
