Aquaporin 5-deficient mouse lungs are hyperresponsive to cholinergic stimulation

Abstract: Although aquaporin 5 (AQP5) is the major water channel expressed in alveolar type I cells in the lung, its actual role in the lung is a matter of considerable speculation. By using immunohistochemical staining, we show that AQP5 expression in mouse lung is not restricted to type I cells, but is also detected in alveolar type II cells, and in tracheal and bronchial epithelium. Aqp5 knockout (Aqp5 ؊/؊ ) mice were used to analyze AQP5 function in pulmonary physiology. Compared with Aqp5 ؉/؉ mice, Aqp5 ؊/؊ mice sh… Show more

Consistent with previous findings seen in the lung tissues of mice, rats, pigs, and humans Krane et al 2001;Skowronski 2010;Kreda et al 2001), the AQP5 antibody noticeably stained (Fig. 4U) the apical membrane of type I porcine pulmonary epithelial cells as a positive control.…”

“…Lack of AQP5 decreased rates of salivary secretion in mice. 16,17 AQP5 expression in apical membrane of acini in salivary gland tissue revealed their secretory abilities. Because epithelial cells must be polarized to secrete fluid unidirectionally, the expression of AQP5 at sites of selfassembly in cultured cells revealed their polarization and secretory potential.…”

“…Some examples include analysis of gene knockouts of ion channels 17,20,25,26 , metabotropic receptors 27,28,29,30 , and downstream signaling cascades 31 . In addition, the antigen challenged mouse is frequently used for asthma studies 32 and the in vitro trachea preparation provides a useful assay for changes in contractility that ensues following development of asthma.…”
