Aptamer-Based Carboxyl-Terminated Nanocrystalline Diamond Sensing Arrays for Adenosine Triphosphate Detection

Abstract: Here, we propose simple diamond functionalization by carboxyl termination for adenosine triphosphate (ATP) detection by an aptamer. The high-sensitivity label-free aptamer sensor for ATP detection was fabricated on nanocrystalline diamond (NCD). Carboxyl termination of the NCD surface by vacuum ultraviolet excimer laser and fluorine termination of the background region as a passivated layer were investigated by X-ray photoelectron spectroscopy. Single strand DNA (amide modification) was used as the supporting … Show more

“…The systematic evolution of ligands by exponential enrichment (SELEX) is used to isolate aptamers with a high affinity for a molecular target from approximately 10 12 –10 15 oligonucleotide combinations [ 45 , 46 , 47 ]. In general, the SELEX process consists of three steps: creating a “library”, binding and separation, and amplification.…”

“…Only covalently bonded molecules with high stability remained on the diamond surface. (9,10) The intensity of the circles on the diamond surface increased with the length of hybridization. First, the fluorescence signal occurred during hybridization.…”

“…(7,8) Research into the application of aptamers to the sensing of small molecules has resulted in the development of advanced biosensors. (9)(10)(11)(12)(13)(14)(15) Aptamers are single-stranded DNA or RNA molecules with various shapes owing to their tendency observed in the systematic evolution of ligands by the exponential enrichment (SELEX) process. (16) Hence, they are extremely good for binding with a target with high affinity, selectivity, and specificity.…”

“…Aptamers are oligonucleotide sequences and can be screened by the technology of a systematic evolution of ligands with exponential enrichment (SELEX) in vitro [ 9 ]. The aptamers and the specific target substances can interact with a high affinity and specificity [ 10 , 11 ]. Currently, different aptamers specific for metal ions [ 12 ], small molecules [ 13 ], drugs [ 14 ], proteins [ 15 ] and cells [ 16 ] have been reported.…”
