Applications of the second virial coefficient: protein crystallization and solubility

Abstract: This article begins by highlighting some of the ground-based studies emanating from NASA's Microgravity Protein Crystal Growth (PCG) program. This is followed by a more detailed discussion of the history of and the progress made in one of the NASA-funded PCG investigations involving the use of measured second virial coefficients (Bvalues) as a diagnostic indicator of solution conditions conducive to protein crystallization. A second application of measuredBvalues involves the determination of solution conditio… Show more

“…Candidate vaccines have been tested as monovalent (single virus), bivalent (two viruses), trivalent (three viruses) and tetravalent (all four serotype viruses) vaccines in Thai volunteers. They were found to be safe and immunogenic in both adults and children [ 56 ] . Four serotypes of monovalent live attenuated dengue virus vaccine candidates were tested for immunogenicity, which were then combined into a tetravalent formulation and given to ten volunteers.…”

“…However, it is nonetheless true that an improved understanding of mechanism will likely propel future advances. An example of a potentially enabling avenue of research involves the osmotic second virial coefficient, a dilute solution parameter that has been demonstrated to predict crystallization behavior (a recent review in this series was devoted to this topic; Wilson & Delucas, 2014). Because the second virial coefficient reflects pairwise interactions between protein molecules (or between protein-detergent complexes), it provides a window into the molecular interactions driving assembly of the crystal lattice.…”
