Applications of Multicomponent Reactions to the Synthesis of Diverse Heterocyclic Scaffolds

Abstract: The sequencing of multicomponent reactions (MCRs) and subsequent cyclization reactions is a powerful stratagem for the rapid synthesis of diverse heterocyclic scaffolds. The optimal MCR is sufficiently flexible that it can be employed to generate adducts bearing a variety of functional groups that may then be selectively paired to enable different cyclization manifolds, thereby leading to a diverse collection of products. The growing interest in diversity-oriented synthesis has led to increased attention to th… Show more

“…17 Nowadays, multi-component reactions (MCRs), sometimes referred to as a "Multicomponent Assembly Process" (MCAP), have attracted enormous attention and have stirred up the interest of synthetic organic chemists due to their well-established advantages and merits. 18 In multi-component reactions whereby more than two reactants combine in a sequential fashion affording highly selective products that retain majority of the atoms of the starting materials. Thus, MCRs are frequently considered as the first choice by synthetic organic chemists for the generation of molecular diversity.…”

“…Multi-component reactions (MCRs) are powerful and valuable tools for the rapid and efficient synthesis of a wide variety of organic molecules. 10,11 In the past few decades, the synthesis of nitrogen-containing heterocyclic compounds has gained prominence as they are widespread in nature. 12 Among a large variety of N-containing heterocyclic compounds, heterocycles containing hydrazine moiety as 'fusion site' have received considerable attention due to their pharmacological properties and clinical applications.…”
