Application of the partial 16S rDNA sequence as an index for rapid identification of species in the genus Bacillus.

Abstract: A comparison of 16S rRNA gene (rDNA) sequences was made among type strains of 69 Bacillus species approved in the International Journal of Systematic Bacteriology (IJSB) until 1998. The results suggested that 5Ј end region (approx. 275 bp) was the hypervariant region (HV region) in the gene and was highly specific for each type strain. Furthermore, a sequence analysis of the HV region of Bacillus strains revealed that this region was highly conserved within the species. These results indicate that the HV regio… Show more

“…Sequence analysis was performed as described previously by Heyrman & Swings (2001). For partial sequencing, two primers were used (reverse 358-339 and reverse 536-519; Heyrman & Swings, 2001) to obtain the first 400-500 bp of the 16S rRNA gene, which has been described as the hypervariable region for the genus Bacillus (Goto et al, 2000). A phylogenetic tree was constructed using BioNumerics 2.0 by applying the neighbour-joining method of Saitou & Nei (1987) on a multiple alignment similarity matrix.…”

“…For the other isolates, only the first 400-500 bases, corresponding to the hypervariable region in Bacillus (Goto et al, 2000), have been sequenced, except for LMG 20967 and LMG 20968, which have not been subjected to sequence analysis because they share an almost identical (GTG) 5 -PCR pattern with LMG 20964 T . The topology of the phylogenetic tree ( Fig.…”

“…sequences of the 16S rRNA gene (Goto et al, 2000). The aim of the present study was to resolve the taxonomic positions of these eight isolates as well as unidentified strain DSM 1857 (Gräf & Bauer, 1973), isolated from a tapwater system, using a combination of taxonomic approaches.…”

“…A large number of aerobic or facultatively anaerobic, rod-shaped, endosporeforming bacteria, many of which belong to the genus Bacillus, are widely distributed in the environment. They are classified as acidophilic, alkalophilic, thermophilic or others according to their nature 3 . Thermophilic Bacillus species, with an optimal growth temperature lying between 45°C and 70°C, have been isolated from a wide range of environments 2 , and the crucial features associated with heat-tolerance have been widely documented.…”
