Application of pharmacokinetic/pharmacodynamic and stochastic modelling to 6‐mercaptopurine micronucleus induction in mouse bone marrow erythrocytes

Abstract: This study investigates the kinetics of bone marrow micronucleated polychromatic erythrocytes and some mechanistic aspects of micronuclei induction using mathematical models. Female mice were administered a single intraperitoneal injection of the purine antagonist 6-mercaptopurine at 50 mg kg(-1). The time course evolution of the drug concentrations in the plasma and the micronucleated polychromatic erythrocyte kinetic rate in bone marrow were observed. Two mathematical models were developed for this study. Th… Show more

“…Published murine pharmacokinetic (PK) models were used to predict drug concentrations of LSD1i and 6MP over time under any given regimen ( Mohammad et al., 2015 ) ( Klés et al., 2003 ). The LSD1i PK model relied on a one-compartment model that starts at the C max with fast absorption kinetics.…”

“…The heart of N. lasiurus fetuses at gestational day 20.5 is similar to the guinea pig [15]. The authors noted the layers of the heart wall, endocardium, myocardium, and epicardium and the structure of the muscle fibers, making it possible to examine similarities between the two species [1].…”

“…In science, rodents are one of the most important groups of experimental animals and have contributed significantly to the advancement of many areas of knowledge and health. The contributions of rodents range from the interpretation of the effects caused by drugs in the body and drug testing [1], to advances in molecular and genetic knowledge [2], to the development of knockout models for various genes and diseases [3]. Currently, knowledge of the embryotoxic effects of drugs and environmental factors such as pollution on embryonic and fetal development has gained special attention with the use of murine models, which allowed the elucidation of factors such as placental changes that lead to disturbances in the development of the uterus during pregnancy and consequently to changes in the normal pattern of fetal growth [4][5][6].…”

“…Stochastic fluctuations in pharmacokinetics and pharmacodynamics have drawn the attention of several research groups. [21][22][23][24] The use of the stochastic approach to address ÔÔmacroÕÕ and ÔÔobservableÕÕ variability problems in population pharmacokinetics and pharmacodynamics has been well demonstrated. [22][23] However, few reports use the stochastic approach to investigate the ÔÔmicroÕÕ variability in pharmacogenomics, probably owing to a limited data resource.…”
