Application of Experimental Design in Preparation of Nanoliposomes Containing Hyaluronidase

Abstract: Hyaluronidase is an enzyme that catalyzes breakdown of hyaluronic acid. This property is utilized for hypodermoclysis and for treating extravasation injury. Hyaluronidase is further studied for possible application as an adjuvant for increasing the efficacy of other drugs. Development of suitable carrier system for hyaluronidase would help in coadministration of other drugs. In the present study, the hyaluronidase was encapsulated in liposomes. The effect of variables, namely, phosphatidylcholine (PC), cholest… Show more

“…All the cell line studies involving coadministration of hyaluronidase along with standard anticancer drug involve direct treatment of the cells with hyaluronidase. However during preclinical and clinical studies it would be easier to control pharmaceutical properties such as drug release and site specificity if hyaluronidase is given using a carrier system [ 5 ]. Among the various polymers used as drug carriers poly(lactic-co-glycolic) acid (PLGA) is widely used due to its biocompatibility [ 6 ].…”

“…The design of experiment (DoE) is an efficient methodology usually adopted to determine the effects of multiple variables on product properties. This approach allows for simultaneous variation of all input parameters rather than assessing the effect of each one on desirable outcomes per time, permitting analysis of their individual and interaction effect on measured responses (41)(42)(43). DoE permits the use of statistical tools such as response surface methodology (RSM) for a rapid, cost effective, and accurate assessment of effects towards optimizing the conditions to achieve desirable product qualities and process performance (44,45).…”

“…Rather than assessing the effect of each input variable on desirable outcomes per time, the Design of Experiment (DoE) is an efficient methodology usually adopted to simultaneously determine the effects of multiple variables on product properties. (CZITROM, 1999;JAIN et al, 2015;KASINATHAN;VOLETY;JOSYULA, 2014). This way, products with optimal physicochemical and biological properties can be prepared in a rapid, cost effective and reproducible manner (BAN et al, 2017;KURHAJEC et al, 2017).…”
