Application of computer‐aided sperm analysis (CASA) for detecting sperm‐immobilizing antibody

Abstract: Using the novel method, manually counting the number of motile spermatozoa becomes unnecessary. The novel method presented here will increase the objectivity and convenience of using the SIT as a clinical indicator.

“…Recently, our group applied the computer‐aided sperm analysis (CASA) for detecting sperm‐immobilizing antibodies as these tests have been carried out by counting the number of motile sperm under a microscope, suggesting that the results may vary because of subjective judgement of the operator, as seen with routine semen examination 20 . We developed a novel method using CASA, and the results were compared with those obtained by the traditional method.…”

“…To manage this, we recently outlined a new method using CASA to make SIT evaluation more convenient and objective as compared to the traditional method 11 . We found the assay results for detecting the SI antibodies and estimating the quantitative titer of SI antibodies to be significantly correlated between the traditional method of manually counting sperm by eye estimation by a highly skilled embryologist and the new method of counting sperm through the chamber slides using CASA 11 . Moreover, this study provided evidence that a highly skilled embryologist is not necessarily required in order to perform this test.…”

“…Over the decades, the CASA system has developed protocols to efficiently analyze the kinematic parameters of sperm in animals [ 98 , 99 ]. However, its use is not just limited to kinematic parameters, it can also analyze morphology, viability, DNA fragmentation, and acrosome reaction [ 100 ], which are considered vital for fertility determination.…”
