AimTo assess the usefulness of a DNA-based method for identifying mushroom species for application in forensic laboratory practice.MethodsTwo hundred twenty-one samples of clinical forensic material (dried mushrooms, food remains, stomach contents, feces, etc) were analyzed. ITS2 region of nuclear ribosomal DNA (nrDNA) was sequenced and the sequences were compared with reference sequences collected from the National Center for Biotechnology Information gene bank (GenBank). Sporological identification of mushrooms was also performed for 57 samples of clinical material.ResultsOf 221 samples, positive sequencing results were obtained for 152 (69%). The highest percentage of positive results was obtained for samples of dried mushrooms (96%) and food remains (91%). Comparison with GenBank sequences enabled identification of all samples at least at the genus level. Most samples (90%) were identified at the level of species or a group of closely related species. Sporological and molecular identification were consistent at the level of species or genus for 30% of analyzed samples.ConclusionMolecular analysis identified a larger number of species than sporological method. It proved to be suitable for analysis of evidential material (dried hallucinogenic mushrooms) in forensic genetic laboratories as well as to complement classical methods in the analysis of clinical material.

Forensic dentistry deals with proper handling, examination, and evaluation of dental records, which are then presented in the interest of law for justice. It plays a major role in identification of deceased individuals who cannot be identified visually or by other means after mass disasters or crimes. Digital forensics has revolutionized the traditional forensic investigations in terms of acquisition, analysis, and reporting of forensic evidence and its application is becoming common in the mass disasters, earthquakes, and terrorism. Sophistication of software and advent of digital technologies such as computers, computer-aided design computer-aided manufacturing systems, digital records, facial reconstruction, touch-free autopsy, and virtopsy has resulted in quick identification and extraction of a large amount of data with reduced sampling bias. This paper focuses on the evolution of forensic dentistry for effective detection and resolution of medico-legal matters and also highlights the use of comparison microscopes and new robotic tools by few forensic laboratories for automation of deoxyribonucleic acid sampling processes for human identification.

Genetyka sądowa jest dynamicznie rozwijającą się dyscypliną naukową. Genetyczna identyfikacja człowieka prowadzona jest z zastosowaniem kompleksowych rozwiązań, które zapewniają wysoką czułość analizy, eliminują niekorzystny wpływ inhibicji i degradacji śladów biologicznych, a także dostarczają maksymalnej liczby informacji przydatnych dla sądu. Ostatnio uwydatniły się jednak problemy, które wcześniej miały mniejsze znaczenie, jak transfer wtórny, mieszaniny DNA czy niekompletność profili DNA. Wydaje się, że potencjał narodowej bazy danych DNA w Polsce wciąż nie jest należycie wykorzystywany i konieczna jest odpowiednia polityka informacyjna w tym względzie. Do nowych metod, które można wykorzystać na poziomie prowadzonego dochodzenia, należą: określanie pochodzenia biogeograficznego, predykcja cech fizycznych, a także przybliżanie chronologicznego wieku człowieka. Metoda sekwencjonowania następnej generacji może już wkrótce całkowicie wyeliminować zastosowanie elektroforezy kapilarnej w genetyce sądowej. Konieczne są dalsze prace umożliwiające wdrożenie jednolitych standardów w interpretacji i ocenie dowodów pochodzących z badania DNA. Utrzymanie odpowiednio wysokich standardów wymaga ustawicznego szkolenia ekspertów oraz właściwego informowania i szkolenia odbiorców ekspertyz.
