Cassava is the staple food for hundreds of millions of people in Africa but its cultivation is seriously constrained by cassava mosaic disease (CMD) in Madagascar, and in Africa in general. This study identified the cassava mosaic geminiviruses (CMGs) involved in CMD in Madagascar and their associated epidemiological characteristics from countrywide surveys. Molecular characterization of CMGs in Madagascar revealed an unprecedented diversity and co-occurrence of six viruses: African cassava mosaic virus (ACMV), East African cassava mosaic Cameroon virus (EACMCV), East African cassava mosaic Kenya virus (EACMKV), East African cassava mosaic virus (EACMV), South African cassava mosaic virus (SACMV) and the recently described Cassava mosaic Madagascar virus (CMMGV). Distinct geographical distributions were observed for the six viruses. While ACMV was more prevalent in the central highlands, EACMV and EACMKV were prevalent in lowlands and coastal regions. Both EACMCV and SACMV occurred in almost all the localities visited. PCR diagnosis revealed that mixed infection (up to four co-infected viruses) occurred in 21% of the samples and were associated with higher symptom severity scores. Pairwise comparisons of virus associations showed that EACMCV was found in mixed infections more often than expected while ACMV and SACMV were mostly found in single infections. A greater abundance of whiteflies was observed in lowland and coastal areas. Nevertheless, infected cuttings remain the primary source of CMD propagation (95%) in Madagascar. (Résumé d'auteur

Cassava mosaic disease (CMD), arguably the most important viral disease of cassava, an important food security crop has in recent times, been reported to be associated with satellites in some parts of sub-Saharan Africa. We investigated the status of Cassava mosaic begomovirus and associated satellites in CMD-affected cassava plants in Ghana using polymerase chain reaction. In all, 110 CMD-affected cassava leaf samples collected from cassava fields in Southern Ghana. Africa cassava mosaic virus (ACMV) was detected in 107 (97%) cassava leaf samples, out of which 76 (69%) was in mixed infections with East Africa cassava mosaic virus-Cameroon (EACMV-CM). EACMV (CM) alone was detected in three (3%) cassava leaf samples. Out of the 110 cassava leaf samples affected by cassava mosaic begomoviruses, satIII and satII were detected in 66 (60%) and 47 (43%), respectively. To the best of our knowledge, detection of CMBassociated satellites in Ghana is the first in the country and possibly in West Africa, and has serious epidemiological implications on the management of CMD, thereby requiring further studies and concerted efforts to safeguard cassava production.

a b s t r a c tDifferent PCR protocols have been established for detection of European fruit trees phytoplasmas; however the majority of the procedures for extracting phytoplasma DNA are complex, time consuming, and expensive, with a risk of contamination or loss of target DNA. In present study, a crude extract preparation method previously used to detect other plant pathogens was adapted to samples from apple trees infected by 'Candidatus Phytoplasma mali'. End-point and real-time PCR detection of 'Ca. P. mali' were used to compare this extraction procedure with an established method for efficient extraction of purified DNA. The crude extract proved fully adequate for phytoplasma detection in samples from 86 in vitro and 35 in vivo apple shoots or plants and 10 periwinkle plants. High inter-and intra-run reproducibility was obtained for phytoplasma detection with different TaqMan MGB-or SYBR Green-based real-time PCR protocols applied to the crude extracts. Real-time PCR applied to serially diluted crude and purified extracts revealed the same phytoplasma detection limit (dilution up to 10 5 ). All results confirm the suitability of this simple, quick, efficient extraction technique for accurate detection of 'Ca. P. mali' in different types of apple and periwinkle samples.
