Application of a Simplified Molecular Protocol to Reveal Mixed Infections with Begomoviruses in Cassava

Abstract: Samples of cassava leaves exhibiting severe symptoms of cassava mosaic disease (CMD) were collected with the PhytoPASS kit in fields surrounding the city of Bujumbura (Burundi). These materials were then sent to Belgium for polymerase chain reaction determination of the CMD begomoviruses inducing the observed symptoms. Different pairs of specific primers were used to amplify DNA sequences specific to African cassava mosaic virus (ACMV), East African cassava mosaic virus (EACMV), East African cassava mosaic Cam… Show more

“…The occurrence of double infections by CMGs has been reported in several studies in Africa: infection of ACMV and EACMCV in Cameroon, the Ivory Coast and Nigeria (Fondong et al ., ; Pita et al ., ; Ariyo et al ., ) and infections by ACMV and EACMV‐UG in Uganda (Pita et al ., ). Triple infection by ACMV, EACMV and EACMV‐UG has been reported in Burundi (Busogoro et al ., ). To the authors' knowledge, this is the first description of mixed infection involving four CMGs in cassava, reflecting the importance of CMD constraint on cassava in Madagascar.…”

“…The Mixed infection of ACMV and EACMV (CM) as revealed in the present study concurs with that of other parts of Africa in which more the one CMB species were reported from CMD-affected cassava plants. (Adjata et al, 2009;Alabi et al, 2007;Busogoro et al, 2008;Ogbe et al, 2006;Were et al, 2004).…”

“…In the present study, an extraction buffer newly developed by the Plant Pathology Department of the Gembloux Agriculture University and used previously for detection of plant viruses (Busogoro et al, 2008;Massart et al, 2008) was tested for 'Ca. P. mali' detection.…”

“…Further research will focus on evaluating its applicability to other phytoplasmas infecting different plant and insect species. As the DNAlis Company has commercialized this extraction buffer with a simplified sampling system (named PhytoPASS, Busogoro et al, 2008), efforts will also be devoted to testing the reliability of this crude extraction method for phytoplasma detection when this simplified sampling system is used.…”
