Application of a modified minicolumn to detection of aflatoxins in corn

Abstract: A simple, rapid and economical minicolumn packed with "lahar" (a local adsorbent material) and Florisil, previously developed for aflatoxins in copra meal, was adopted for corn. at the detection limit of 5 ƒÊg/kg of aflatoxin.

“…Mycotoxin Analysis. For the analysis of aflatoxins, all corn samples were qualitatively screened for aflatoxins according to the minicolumn method described previously (14). The positive minicolumn response for samples containing 5 µg/kg total aflatoxins and above was determined by visualizing the column under long-wave UV light.…”

“…Solid-phase extraction (SPE) has been used in cleanup procedures saving a lot of time, being solvent-efficient and economical [7,37]. The use of SPE cartridges is simple and usually involves three steps: loading the sample, washing the impurities and eluting the toxin to be determined [13].…”

“…The use of SPE cartridges is simple and usually involves three steps: loading the sample, washing the impurities and eluting the toxin to be determined [13]. A variety of solid phase have been used in aflatoxin analysis, such as silicagel, surfacedmodified bonded silica like florisil and octadecil, a mixture of reversed-phase, ion exclusion, and ion exchange adsorbents (ISOLUTE and ROMER multifunctional column) and other minicolumns [1,3,6,7,25,27].…”

“…Mycotoxin contamination in the agricultural products of the country happens during the pre-and post-harvest stages of production (27). However, research in the Philippines is more focused on aflatoxin contamination in food and feeds because of its prevalence and the serious threat to human and animal health (29,30). According to Arim et al (29), more than 50% of corn stocks in government and privately owned warehouses were estimated to be contaminated with non-tolerable aflatoxin concentrations ( > 20 μg/kg).…”
