Application and Prospect of CRISPR/Cas9 Technology in Reversing Drug Resistance of Non-Small Cell Lung Cancer

Abstract: Cancer drug resistance has always been a major factor affecting the treatment of non-small cell lung cancer, which reduces the quality of life of patients. The clustered regularly interspaced short palindromic repeats/CRISPR associated protein 9 (CRISPR/Cas9) technology, as an efficient and convenient new gene-editing technology, has provided a lot of help to the clinic and accelerated the research of cancer and drug resistance. In this review, we introduce the mechanisms of drug resistance in non-small cell l… Show more

“…Physical methods including electroporation and microinjection are regularly used to delivery CRISPR components in vivo studies. Microinjection has the limitations of high costs and poor efficiency (Huang et al, 2022). However, an electroporation-based strategy transports Cas9/sgRNA ribonucleoproteins (RNPs) into mouse fertilized eggs with 100% efficiency, realizing extremely efficient gene editing in vivo (Chen et al, 2016).…”

“…Due to their efficient infection ability towards the lung epithelium in adult mice, ADVs are regarded as an ideal method for constructing the aforementioned lung cancer model harboring EML4-ALK inversion (Maddalo et al, 2014). Adenoassociated viruses (AAVs) are less immunogenic compared to other viral vectors, exhibiting clinical therapeutic promise (Huang et al, 2022).…”

“…Interestingly, targeted genetic changes linked to resistance mechanisms have been identified by CRISPR/Cas9 gene editing, which shows potential for treating drug resistance in respiratory diseases [ 158 ]. Clinical trials are underway to evaluate the safety and efficacy of CRISPR/Cas9 gene editing in overcoming drug resistance in respiratory disorders.…”

“…Lung cancer has been identified as the second most diagnosed type of tumor, causing 1.8 million cancer-induced deaths [ 207 ]. Since cancer drug resistance has been the main culprit in providing relief to lung cancer patients, CRISPR techniques have offered a glimpse of hope for a better quality of life [ 206 ].…”
