Apoptin induces pyroptosis of colorectal cancer cells via the GSDME-dependent pathway

Abstract: Apoptin is a small molecular weight protein encoded by the VP3 gene of chicken anemia virus (CAV). It can induce apoptosis of tumor cells and play anti-tumorigenic functions. In this study, we identified a time-dependent inhibitory role of apoptin on the viability of HCT116 cells. We also demonstrated that apoptin induces pyroptosis through cleaved caspase 3, and with a concomitant cleavage of gasdermin E (GSDME) rather than GSDMD. GSDME knockdown switched the apoptin-induced cell death from pyroptosis to apop… Show more

“…But, the cleaved GSDMD cannot be detected in osthole-induced HeLa cells (supplementary Figure S4 ). Among the alternative pathways, the most widely studied is GSDME-induced pyroptosis through caspase-3 [ 30 ]. GSDME is a member of the gasdermin family which contains a cytotoxic N-terminal domain to form pore and induce membrane permeabilization.…”

“…8). Although FB1 showed pro-apoptotic effects (Hou et al, 2021), the FB1 concentration used was of low-dose as demonstrated by the weight change and lung histopathology, however, low-dose FB1 only upregulated the pro-apoptosis protein Caspase 9 both in vivo and in vitro, recent researches have proved that Caspase 9 is also attributed to pyroptosis activation (Liu et al, 2022), as our results showed, low-dose FB1 induced signi cant in ammatory response and pyroptosis-related protein expressions in mouse lung and A549. Furthermore, our results showed that apoptosis didn't contribute to the FB1-promoted in ammatory response and injury (Fig.…”

“…In the classical pyroptosis pathway, when certain inflammasomes (NLRP1, NLRP3, NLRC4, and AIM2) stimulate pyroptosis-related pattern recognition receptors, they recruit and activate CASP1 either directly or by recruiting the receptor protein ASC recruits and activates CASP1, which successively promotes the maturation of IL-1β and IL-18 precursors and cleaves GSDMD [ 10 ]. Among other pyroptosis pathways, activated CASP4/5 specifically cleaves GSDMD [ 43 ], as well as CASP8 and CASP9 by affecting CASP3, causing cleavage of GSDME [ 44 , 45 ]. Subsequently, pyroptosis pathways mediated by cleavage of other GSDM family proteins were identified, including GSDMA, GSDMB, and GSDMC [ 24 , 46 , 47 ].…”
