Apigenin Induces Apoptosis in Human Leukemia Cells and Exhibits Anti-Leukemic Activity In Vivo

Abstract: In this study, we investigated the functional role of Akt and JNK signaling cascades in apigenin-induced apoptosis in U937 human leukemia cells and anti-leukemic activity of apigenin in vivo. Apigenin-induced apoptosis by inactivation of Akt with a concomitant activation of JNK, Mcl-1 and Bcl-2 down-regulation, cytochrome c release from mitochondria and activation of caspases. Constitutively active myristolated Akt prevented apigenin-induced JNK, caspases activation, and apoptosis. Conversely, LY294002 and a d… Show more

“…In addition to inducing apoptosis in leukemia cells, apigenin-7 glycoside has been shown to induce granulocyte differentiation that was confirmed by the presence of significant amounts of CD11b positive cells [186]. Evidence also suggests that PI3K/Akt and JNK kinases and their related pathways are potential targets for apigenin-induced apoptosis in leukemia cells [187]. Apigenin-induced expressions of α, β, and γ globin genes increased the expression of glycosporin as a marker for differentiation.…”

“…Many of the chemoprevetive effects of apigenin are related to its inhibition of Akt activity. It is well known that PI3K/Akt signaling plays an important role in inhibiting apoptosis for cancer cell survival and growth, and apigenin has been reported in numerous studies in vitro as well as in vivo to enhance apoptosis by inactivation of Akt [113, 114]. Apigenin has also been demonstrated to inhibit ovarian tumor metastasis through down-regulation of MMP-9, which is mediated by Akt signaling [91].…”

“…Apigenin inactivates Akt to trigger apoptosis in human prostate cancer in vitro and in vivo (Kaur et al, 2008). Apigenin also induces apoptosis in human leukemia cells and exhibits anti-leukemic activity in vivo (Budhraja et al, 2012). Apigenin inhibits expression of VEGF and angiogenesis in human lung cancer cells (Liu et al, 2005).…”
