Aortic aneurysm associated with cardiac defects in theophylline stimulated chick embryos

Abstract: Theophylline(1 ml 0.02 M in 0.15 M saline) was administered to embryonic chicks (Hamburger-Hamilton developmental stages 24.27). The drug was topically applied to the surface of the chorioallantoic membrane in the vicinity of the embryonic heart. At necropsy, 63% (44/70) of the theophylline-exposed embryos demonstrated an aneurysm in the ascending aorta. Among the cases exhibiting aortic aneurysm, 48% (21/44) of the embryos demonstrated ventricular septal defects. Four cases of truncus arteriosus were also obs… Show more

“…A broad spectrum of congenital cardiac anomalies in chick embryos has been demonstrated in previous reports [Ishikawa et al, 1978a;Gessner, 1966Gessner, , 1970Stephan, 1952;Harh, 19731. Such experiments have been performed to induce developmental Fig.…”

“…Gilbert et al [1977] and Ishikawa et al [1978] reported that stage 26 chick embryos treated with caffeine or theophylline developed aneurysms of the ascending aorta. We speculate that the aortic aneurysm forms as a result of an increase in blood vessel wall stress.…”

“…43 Theophylline also produces aortic aneurysms in chicks if they are exposed to this agent in utero. [45][46][47] The mechanism of action appears to be through the effect of theophylline on cyclic adenosine 3Ј,5Ј-monophosphate (AMP). Intracellular theophylline increases AMP and is associated with inhibition of cellular mitoses, [48][49] so that theophylline leads to arrested development of the arterial wall.…”
