Gelchromatographische und elektrophoretische Untersuchungen von ultrafiltrierten Molkenproteinenl T. SIENKIEWICZ Die Anwendung von Sephadcx G-75 zur gelchromatographischen Molkcnproteinfraktionierung gestattet bei aufsteigender Chromatographie die Gewinnung vou Immunglobulinen, Serumalbumin, p-Laktoglobulinen, Minorfraktionen dcs a-Laktalbumins und des a-Laktalbumins. Wiederholte Chromatographie an Sephadex G-75 gibt die Miiglichkeit der Auftrennung aller Proteinhauptkornponenten mit guter Auflosung. Die mit dieser Methode untersuchte spruhgetrocknete Ultrafiltrationsprobe der Molkenproteine weist einen erniedrigten Gehalt an is-Laktoglobulinen (45.04%) und einen erhohten Gehalt der hochmolekularen Immunglobulinfraktion (28,55y0) auf. Entsprechende Wertefur die unbehandelte Probe waren: 64.14% bzw. g.4zyo. Die elektrophoretischc Prufung der hochmolekularen Fraktion ergab Proteinbanden mit einer relativen Bewegljchkeit entsprechend der Immunglobulinc, Serumalbumin und 8-Laktoglobulin. Die Polyacrylamidgelelektrophorese von Molkenproteinen unter Zusatz von 8M Harnstoff ergab fur die Untersuchungsprobe eine bessere Auflosung. Der Unterschied der relativen Beweglichkeit von /?-Lactoglobulin und a-Lactalbumin, ausgedriickt in der Differenz der Rp-Wertc, betrug RTff* = o,r7 bei 8 M Harnstoff und RFff. = o,og ohne Harnstoffzugabe.
