Anvillea garcinii extract inhibits the oxidative burst of primary human neutrophils

Abstract: Background Anvillea garcinii Coss. & Durieu (Anv) plant is used as a traditional North African medicine against several diseases associated with inflammation. At inflammatory sites, reactive oxygen species (ROS) produced in excess by activated phagocyte NADPH oxidase (NOX2) can accentuate inflammatory responses. Thus, we investigated if Anv-water soluble polysaccharides could modulate primary human neutrophil oxidative burst in vitro.MethodsHuman neutrophils were isolated from fresh whole blood and O2 .- gener… Show more

“…During PDT, BAP31 is cleaved before the mitochondria changes and mediates the release of ERS and Ca 2+[43] . From the point of view of enzyme catalysis, PDT regulates flavin adenine dinucleotide, ubiquinones, and cytochrome in mitochondrial respiratory chain complexes I and III to produce ROS[ 44 - 46 ]; mitochondrial triphosphopyridine nucleotide(NADPH) oxidase and xanthine oxidase catalyze the production of O 2− [ 47 ]; mitochondrial myeloperoxidase myeloperoxidase (MPO) catalyzes the production of OH; and protein kinase C catalyzes the production of H 2 O 2 [ 48 , 49 ]. Because the dynamic changes in ROS are rapid, in trace amounts, and complex, it is difficult to accurately detect the process by which ROS are produced, from exogenous to endogenous ROS, by PDT within a short period.…”

“…Anti-MPO activity was observed for Anvillea garcinii subsp. radiate ( Boukemara et al, 2016 ). This plant was used for symptomatic treatment of cold ( Perveen et al, 2020 ), but the successful treatment was related to the presence of flavonoids and sesquiterpene lactones, which show significant antibacterial and antifungal properties ( Perveen et al, 2020 ).…”

“…Blood neutrophils were isolated from the athletes at rest 24 hours after the last exercise session. Neutrophils were treated with fMLP (Nformylmethionyl-leucyl-phenylalanine), a chemotactic factor (31)(32)(33) to measure cell migration, PMA (phorbol 12-myristate 13acetate), an activator of PKC (34) to measure ROS production, or LPS (lipopolysaccharide), a toll-like receptor 4 agonist that activates NF-ƙB transcription factor (35,36) to evaluate secretion of cytokines (IL-1b, TNF-a, IL-6, IL-8, and IL-10). The same parameters were measured in unstimulated neutrophils.…”
