Background. Peptic ulcer disease causes significant mortality and morbidity. Plant kingdom provides a useful source for the development of new antiulcer agents. Croton macrostachyus is traditionally used to treat peptic ulcer in Ethiopia. This study aimed to evaluate the antiulcer activity of C. macrostachyus root extracts in rodents using different models. Methods. The crude extract was obtained by cold maceration in 80% methanol and fractionated with chloroform, ethyl acetate, and distilled water. The antiulcer activity was evaluated using pylorus ligation-induced ulcer model in Sprague Dawley rats and acidified ethanol-induced ulcer model in Swiss albino mice. The test groups received three doses (100, 200, and 400 mg/kg) of the crude extract and fractions for 7 days before induction of ulcer. Positive controls received omeprazole 30 mg/kg for the pylorus ligation-induced ulcer model and sucralfate 100 mg/kg for the acidified ethanol-induced ulcer model. Negative controls received vehicle (2% tween 80). Results. The crude hydromethanolic extract of C. macrostachyus showed significant (p<0.05) antiulcer activity on both pyloric ligation and HCl/ethanol-induced ulcer in rats and mice. It has antisecretary effect (p<0.001) as well. All three administered doses of chloroform fraction (p<0.05) and only higher doses of ethyl acetate fraction (p<0.05) possessed significant antiulcer activity. In contrast, the aqueous fraction did not have significant antiulcer effect at all tested doses. Conclusion. The present study demonstrated that the crude extract, chloroform, and ethyl acetate fractions possessed significant dose-dependent antiulcer activity.

Gastroprotection Studies of Schiff Base Zinc (II) Derivative Complex against Acute Superficial Hemorrhagic Mucosal Lesions in Rats

BackgroundThe study was carried out to assess the gastroprotective effect of the zinc (II) complex against ethanol-induced acute hemorrhagic lesions in rats.Methodology/Principal FindingThe animals received their respective pre-treatments dissolved in tween 20 (5% v/v), orally. Ethanol (95% v/v) was orally administrated to induce superficial hemorrhagic mucosal lesions. Omeprazole (5.790×10−5 M/kg) was used as a reference medicine. The pre-treatment with the zinc (II) complex (2.181×10−5 and 4.362×10−5 M/kg) protected the gastric mucosa similar to the reference control. They significantly increased the activity levels of nitric oxide, catalase, superoxide dismutase, glutathione and prostaglandin E2, and decreased the level of malondialdehyde. The histology assessments confirmed the protection through remarkable reduction of mucosal lesions and increased the production of gastric mucosa. Immunohistochemistry and western blot analysis indicated that the complex might induced Hsp70 up-regulation and Bax down-regulation. The complex moderately increased the gastroprotectiveness in fine fettle. The acute toxicity approved the non-toxic characteristic of the complex (<87.241×10−5 M/kg).Conclusion/SignificanceThe gastroprotective effect of the zinc (II) complex was mainly through its antioxidant activity, enzymatic stimulation of prostaglandins E2, and up-regulation of Hsp70. The gastric wall mucus was also a remarkable protective mechanism.

The leaves of Bauhinia monandra prepared as poultices are used in many Brazilian tribes and some Nigerian communities to relieve swelling and pains, arthritis, diabetes among other ailments. These folkloric claimed benefits inspired this study in which topical and systemic anti-inflammatory activities of a methanol extract of B. monandra leaves (MEBmL) was assessed in rodents. The anti-inflammatory activities of B. monandra were determined by egg albumin-induced (systemic) paw edema in rats, and xylene-induced (topical) ear edema in mice. Treatment of rats with MEBmL at different concentration (200 and 400 mg/kg) led to significant (p<0,05) reduction (31.0% and 28.0% respectively) of the egg albumin-induced paw edema. Topical application of mice ear with MEBmL (50, 100 mg/kg) also significantly (p<0.05) reduced (39.50% and 41.87% respectively) xylene-induced ear edema. Based on the findings of the study, B. monandra has a promising therapeutic potential with its anti-inflammatory activities as associated with folkloric use of the plant.
