Antitumor Activity of Pulsatilla koreana Saponins and Their Structure-Activity Relationship

Abstract: Seventeen saponins isolated from the root of Pulsatilla koreana were examined for their in vitro cytotoxic activity against the human solid cancer cell lines, A-549, SK-OV-3, SK-MEL-2, and HCT15, using the SRB assay method, and their in vivo antitumor activity using BDF1 mice bearing Lewis lung carcinoma (LLC). The saponins 5-17, with a free acidic functional group at C-28 of aglycon, exhibited moderate to considerable cytotoxic activity, however, the saponins 1-4, esterified with a trisaccharide at C-28 of ag… Show more

“…5) In addition, a total 17 triterpenoid saponins including six new saponins were isolated from Pullsatilla koreana N. root and the structure-activity relationships of antitumoral saponins was published. 6,7) Among them, Pullsatilla saponin D (1, inhibition ratio (IR): 66.9% against LLC lung carcinoma xengraft model at 6.0 mg/kg/d i.p., ED 50 3.04- 50 1.57-8.36 mM) exhibited highly potent anticancer activity. Pulsatilla saponin D shows five times more active in anticancer test than its aglycone (ED 50 ; Ͼ50 mM).…”

“…1) not only displayed strong in vitro anticancer activities, but also more potent in vivo antitumor activities in mice bearing Lewis lung carcinoma, exceeding the effects of even taxol and doxorubicin. 8) In addition, we reported total synthesis of Pulsatilla saponin D (PSD). 9) These natural saponin products were obtained in small amounts, which was a limiting factor.…”

“…[2][3][4] Oleanane-type triterpenoid saponins, lupane-type triterpenoid saponins, quinones, phenylpropanoids, and flavonoid glycosides have been isolated from the root and aerial parts of P. koreana in previous phytochemical studies. [5][6][7][8][9] However, there have been few studies that have specifically aimed to identify compounds from the flowers of P. Koreana.…”
