Antitumor Activity of Ligustilide Against Ehrlich Solid Carcinoma in Rats via Inhibition of Proliferation and Activation of Autophagy

Afnan F Alshehri,
Ahmed E Khodier,
Mohammed M Al-Gayyar

Abstract: BackgroundCancer is the second-leading cause of death worldwide. According to a 2018 WHO report, 9.6 million deaths occurred globally due to cancer. Ehrlich carcinoma is characterized by rapid proliferation and a short survival time. Ligustilide is a phthalide derivative and is one of the main compounds in Danggui essential oil and Rhizoma Chuanxiong. It has many protective effects, such as anticancer, anti-inflammatory, antioxidant, and neuroprotective effects. AimsWe conducted this study to investigate the a… Show more

“…Notably, LIG promoted apoptosis in ESC by significantly increasing the expression of Bcl2. 29 It also dramatically inhibits the viability and migration of HCC and oral cancer cells. 31 In this paper, we investigated the functions of LIG and NBP in U251 the proliferation, migration, invasion, and apoptosis.…”

“…Emerging evidence indicates that LIG has anti-tumor effects on Ehrlich solid carcinoma (ESC), 29 hepatocellular carcinoma (HCC), 17 prostate cancer, 16 breast cancer 30 and brain tumor, 15 whereas NBP exhibits anti-cancer activity in lung cancer. 19 There are few reports on the therapeutic effects of LIG and NBP, especially NBP, on gliomas.…”

“…This pathway can be activated either directly through elevated levels of ROS and VEGF or indirectly through the release of histamine [16]. Autophagy inhibition is a function of mTOR, which enhances the signaling pathways of MAPK and AKT [8]. Inhibiting PI3K and mTOR has shown promising results in combating tumor cells [6].…”

“…The control group (no ESC nor echinacoside) did not receive any treatment during the experiment. The ESC group (ESC with no echinacoside) was injected with 200 µL of 2x10 6 tumor cell suspension in phosphate buffer saline into the left hind limb using intramuscular injection [6][7][8]. In the ESC+echinacoside group, the first tumor was injected in rats and left to grow for 14 days, the time taken for the tumor to appear.…”
