Antitumor activity of electrospun polylactide nanofibers loaded with 5-fluorouracil and oxaliplatin against colorectal cancer

Abstract: The purpose of this study was to evaluate both in vitro and in vivo anticancer activities against colorectal cancer (CRC) of electrospun polylactide (PLA) nanofibers loaded with 5-fluorouracil (5-Flu) and oxaliplatin. For in vitro evaluation, human CRC HCT8 cells were directly exposed to the drug-loaded fiber mats, followed with MTT and flow cytometry (FCM) assay. For in vivo evaluation, the drug-loaded fiber mats were locally implanted into mouse colorectal CT26 tumor-bearing mice, followed with histological … Show more

“…The cytotoxicity of T-PNPs, T-SLNs, and T-NLCs was tested in U87 MG cells using the MTT assay (Zhang et al, 2016). Briefly, cells were seeded in a 96-well plate at a density of 1 Â 10 4 cells/well and allowed to adhere for 24 h prior to the assay.…”

“…The cytotoxicity of 5-FU (1 µg/ml) and oxaliplatin (l-OHP) (2 µg/ml) (both Shanghai Lichen Biotechnology Co., Ltd.) were determined using the MTT assay-based Cell Growth Determination kit (Sigma-Aldrich; Merck Millipore), as described previously (12). Briefly, cells were cultured in a flat 96-well plate at a density of 1,000-10,000 cells/well until ~80% confluence was reached.…”

“…Electrospinning is a technique mostly used for producing polymeric nano-or micro-fibers. The advantage of this method is that structure and morphology of the fiber can be controlled, by modifying several parameters such as polymer type, concentration of solution, solvent or electrospinning conditions (Zhang et al, 2011;Zhang et al, 2016). Two nozzle electrospinning technique was used in this study to provide simultaneous spinning of fiber polymer and CsAloaded nanoparticles to obtain nanoparticle-loaded fibers (Pu et al, 2014).…”
