Antithrombin Reduction Corrected Thrombin Generation in Samples from Hemophilia A and B Patients with Inhibitors

Abstract: Background: Severe hemophilia A and B patients with inhibitors experience serious musculoskeletal hemorrhage as well as high risk of limb and life threatening bleeds. However, lack of effect of FVIII or FIX substitution therapy and short functional half-life of by-passing agents, leave these patients with very limited bleed preventive treatment options. ALN-AT3 (Alnylam Pharmaceuticals, Cambridge, MA, USA), a subcutaneously administered investigational RNAi therapeutic targeting reduction of antithrombin for p… Show more

“…58,59 Furthermore, 50 and 90% antithrombin reduction in 12 HA and HB inhibitor plasmas showed increased peak TG without overcorrection beyond values observed in normal controls. 60 Large clinical studies in inhibitor and noninhibitor patients are being conducted aiming to assess safety better and provide guidance about appropriate dosing regimen for factor/bypassing agents replacement in the setting of bleeding and surgery.…”
