Antithrombin III (AT3) polymorphism in the marsupialMonodelphis domestica: Identification and genetics

Abstract: Antithrombin III polymorphism was observed in the gray short-tailed opossum, Monodelphis domestica, by either one-dimensional polyacrylamide gel electrophoresis (PAGE; pH 7.9), two-dimensional PAGE (agarose, pH 5.4; 12% T, pH 7.9), or isoelectric focusing (pH 4.2-4.9) followed by immunoblotting with rabbit antiserum to human antithrombin III. Family studies demonstrated an inheritance of three codominant autosomal alleles, AT3A, AT3B, and AT3C, and a population study revealed frequencies of 0.70, 0.10, and 0.2… Show more

“…Proteins were electroblotted from stained (for TF) and unstained (for ES) ID-PAGE gels on to nitrocellulose (0.45 pm) in a Bio-Rad Trans Blot cell at 3 mA/cm2 for 1 2 min and probed with either rabbit antiserum to human TF (Dako) (1:500 dilution) or equine esterase (1:20 dilution) using the procedure described in the Bio-Rad bulletin 1721 (goat anti-rabbit IgG alkaline phosphatase conjugate kit). Plasmas (5pl) were also subjected to polyacrylamide isoelectric focusing (PAIEF), 5%T pH4.2-4.9 (Pharmalyte) with 3% N-(2-acetamido)-Z-aminoethane sulphonic acid 408 (ACES; BDH chemicals) (gel dimensions Bell, Arthur, Breen 240xllOx0.5mm) as described by Bell et al (1992) for 3 h at constant 8 W and 15°C and stained for ES activity as described for the I D -PAGE gels. Plasmas (5pl) were also subjected to polyacrylamide isoelectric focusing (PAIEF), 5%T pH4.2-4.9 (Pharmalyte) with 3% N-(2-acetamido)-Z-aminoethane sulphonic acid 408 (ACES; BDH chemicals) (gel dimensions Bell, Arthur, Breen 240xllOx0.5mm) as described by Bell et al (1992) for 3 h at constant 8 W and 15°C and stained for ES activity as described for the I D -PAGE gels.…”
