The presence of antisperm antibodies might impair fertilization by affecting sperm function in different ways. The aim of this study was to analyze the incidence of antisperm antibodies and evaluate its impact on sperm movement, in normozoospermic and abnormal semen samples. Samples from 144 patients were classified as normozoospermic or abnormal, according to WHO guidelines and Kruger`s strict criteria. Motilility was assessed by a computer-assisted system by placing a drop of semen in a Makler chamber. Antisperm antibodies were detected using the Mar Test and Immunobead tests to identify immunoglobulin location and isotype. Results showed no association between the presence of antisperm antibodies and semen quality. Antibody incidence was 15 and 9.5% for normozoospermic and abnormal semen samples, respectively. Motion analysis showed that, although sperm linearity was significantly reduced in abnormal patients with antibodies, the existence of an immune response did not affect the sperm pathway in most cases. This study suggests that sperm motion analysis would not be of great assistance in the screening of an immunological male fertility factor.

This investigation was designed to study the effect of two different protocols on the indirect sperm-associated antibody test on serum performed using Bio-Rad immunobead (IBT) at two andrology laboratories. Aliquots of 31 serum samples from infertile couples were analyzed by both protocols. The IBT was negative by both protocols (100% concordance) for 18 serum samples. Seven of the 13 remaining samples were positive by both protocols (> or = 10% bead attachment by protocol A and > or = 20% bead attachment by protocol B), for a concordance of 54%. The remaining six samples were positive by one of the two protocols. The overall concordance for positive and negative results was 81% (25 of 31 samples). Protocol A detected a higher percentage of bead attachments for IgG and IgA, while protocol B detected a higher percentage of bead attachments for IgM. The discordance in the results of IgA attachment obtained by the two protocols was statistically significant (p < .05). A standardized, uniform protocol for the indirect IBT is needed.

A simple solid‐phase indirect immunogold assay for light microscopic localization of the binding of sperm‐reactive antibodies in human sera to surface‐associated antigenic sites on human sperm is described. Antigen‐antibody interaction is localized by using a gold‐tagged second antibody directed against the first antibody. Silver enhancement of the 5 nm gold particles makes the immunologic reaction visible by light microscopy. An ELISA was used to identify serum samples as positive or negative for sperm‐reactive antibodies. These serum samples were blind‐coded and were used to develop and to evaluate this solid‐phase indirect immunogold assay. When results obtained by this immunogold assay for the detection of sperm‐reactive antibodies were compared with those obtained by the ELISA, there was agreement in 82 of the 100 sera tested. As a localization assay, this indirect immunogold assay is simple to perform and enables stable, high resolution light microscopic localization of the pattern of antigenic sites on sperm surface membranes.
