Enriched populations of oligodendrocytes were isolated from adult human brains of 3-15 hours postmortem using the trypsinin digestion-Percoll density gradient method and were cultured for an extended period of time up to 6 months. Cell type specific antigens that were expressed by oligodendrocytes were galactocerebroside, myelin basic protein, proteolipid protein, 2'3'-cyclic nucleotide 3'-phosphohydrolase and myelin-associated glycoprotein. In addition, HLA-A,B,C and HLA-DR, respectively, Class I and Class II antigens of the major histocompatibility complex, were demonstrated on oligodendrocytes. Three classes of gangliosides, GM1, GM4, and GD3, were also demonstrated on oligodendrocytes, while GM1 and GM4 gangliosides were detected on the surface of astrocytes. The presence of "transitional" or "bipotential" glial cells that were derived from oligodendrocytes and that expressed both oligodendroglial and astrocytic phenotypes was demonstrated. Treatment of the cells by cyclic AMP and its derivatives reversed this dual phenotypic expression back to the oligodendroglial trait. Electron microscopic examination of oligodendrocytes indicated that they were capable of synthesizing and assembling myelin sheaths in culture in the absence of any neuronal signal input.

Suspensions of bovine oligodendrocytes were used to study the immunofluorescent staining properties of sera from multiple sclerosis (MS) patients and normal individuals. All sera (14 MS patients, 8 patients with other neurological diseases, and 11 normal individuals) showed positive oligodendrocyte staining by indirect immunofluorescence. Staining persisted after extensive absorption of sera with bovine liver to remove nonspecific binding. Similar findings were obtained for cerebrospinal fluid from all 5 MS patients as well as all 5 patients with other neurological diseases. In additional studies to determine if binding is mediated by the Fc fragment of IgG, results were: (1) ultracentrifuged normal and MS sera failed to react with oligodendrocytes, (2) positive staining with oligodendrocytes was observed after heat aggregation of IgG, (3) ox red blood cells, complexed with antibody, reacted with oligodendrocytes to produce strong rosette formation, and (4) the rosette formation could be blocked by prior treatment of oligodendrocytes with heat-aggregated IgG. The studies fail to support a previous claim for specific binding of immunoglobulins to oligodendrocytes in MS. However, they confirm and extend previous findings that the binding of IgG to oligodendrocytes may not necessarily be an antigen-antibody reaction. Therefore, adherence reactions with a putative antibody must exclude Fc-mediated binding.

A marker for oligodendrocytes and its relation to myelinogenesis: An immunocytochemical study with experimental allergic encephalomyelitis serum and C.N.S. cultures

To investigate a possible marker for oligodendrocytes and its relation to myelinogenesis, experimental allergic encephalomyelitis (EAE) serum has been used to study C.N.S. cultures from the time of explantation to maturity at 26 days in vitro (DIV). Cultures of foetal mouse spinal cord were exposed for 1 h to heated (complement-inactivated), rabbit anti-bovine white matter (WM-EAE) or control serum, fixed and processed by an immunoperoxidase technique for demonstrating bound immunoglobulin (Ig) by light and electron microscopy. From 5 to 26 DIV, cells morphologically identical to oligodendrocytes displayed binding of Ig to the plasmalemma of the cell body and its processes. At 5 DIV, immunoreactive oligodendrocytes had a large nucleus and nucleolus, prominent Golgi apparatus, and microtubules but no filaments. Occasionally a centriole was present, suggesting an early stage of differentiation. In myelinated cultures (from 11-12 DIV onwards), reaction product was present on the oligodendroglial outer plasmalemma apposed to myelin and along the outer loop. Sometimes it extended into the external mesaxon, outer layer of myelin, inner mesaxon and periaxonal space. No other structures were reactive, and oligodendroglia did not bind control Ig. These findings indicate that WM-EAE serum can be used as a marker for oligodendrocytes in cultures from 5 DIV onwards. The findings that oligodendrocytes acquire the antigen(s) prior to myelination and that the antigen(s) is localized on the plasmalemma of the inner and outer loops of actively myelinating oligodendroglial processes suggest that the antigen(s) may have a role in oligodendrocyte maturation and myelinogenesis. The antigen(s) involved is not yet established, but it is probably not myelin basic protein. This marker should prove useful in studies of C.N.S. development and the demyelinating diseases.
