Antisense Transcription in the Mammalian Transcriptome

Abstract: Antisense transcription (transcription from the opposite strand to a protein-coding or sense strand) has been ascribed roles in gene regulation involving degradation of the corresponding sense transcripts (RNA interference), as well as gene silencing at the chromatin level. Global transcriptome analysis provides evidence that a large proportion of the genome can produce transcripts from both strands, and that antisense transcripts commonly link neighboring "genes" in complex loci into chains of linked transcri… Show more

“…This represents a negative feedback loop, in which aHIF inhibits the translation of HIF-1a mRNA after chronic hypoxia (Uchida et al 2004). A 20% of the human genome consists of these naturally occurring antisense transcripts, which regulate gene transcription, RNA splicing, polyadenylation, editing, stability, transport, and translation (Yelin et al 2003, Chen et al 2004, Katayama et al 2005. Importantly, besides acting as a transcription factor for hypoxia-inducible genes, HIF-1a also stabilizes the tumor suppressor gene p53 under early hypoxic conditions (An et al 1998).…”
