Aim: To investigate the effects of wogonin (5,7-dihydroxy-8-methoxyflavone) extracted from Scutellaria baicalensis Georgi (S baicalensis) on lipotoxicity-induced apoptosis of vascular smooth muscle cells (VSMCs) and the underlying mechanisms. Methods: Cultured VSMCs were used. Apoptosis of VSMCs was induced by palmitate (0.75 mmol/L), and detected using TUNEL assay. The expression levels of protein and phosphorylated protein were measured using Western blot analysis. Results: Treatment of VSMCs with wogonin (10, 25 and 50 µmol/L) significantly attenuated the apoptosis and endoplasmic reticulum (ER) stress induced by palmitate in concentration-and time-dependent manners. Wogonin (50 µmol/L) decreased palmitate-induced reactive oxygen species (ROS) generation. The ER stress inhibitor 4-phenyl butyric acid (5 mmol/L) significantly decreased palmitateinduced apoptotic cells, and occluded the anti-apoptotic effect of wogonin (25 µmol/L). Wogonin (10, 25 and 50 µmol/L) significantly reduced the intracellular diacylglycerol (DAG) accumulation and expression levels of phosphorylated PKCs in palmitate-treated VSMCs. Conclusion: Our results suggest that wogonin inhibits lipotoxicity-induced apoptosis of VSMCs via suppressing the intracellular DAG accumulation and subsequent inhibition of PKC phosphorylation. Wogonin has therapeutic potential for the prevention and treatment of atherosclerosis.

1) These compounds might contribute to the anti-hypertensive effect collectively. However, the anti-hypertensive effect of Eucommia ulmoides OLIV. bark remains largely unclear.Pharmacological studies have revealed that Eucommia ulmoides OLIV. bark extract induce endothelium and NO-cyclic guanosine monophosphate (cGMP) dependent relaxation in the rat thoracic aorta.2) Another report demonstrated that the endothelium-dependent vascular relaxation induced by the bark extract is mediated by NO and endothelium-derived hyperpolarizing factor in small vessels.3) However, the vasorelaxing components have been unclear. Recently, we found oroxylin A and wogonin isolated from Eucommia ulmoides OLIV. bark could significantly lower the perfusion pressure.4) In the previous study, we had reported that oroxylin A could relax rat thoracic aorta and it was endothelium and NO dependent.5)The present study was undertaken to investigate vasodilatory effect of wogonin and its mechanism.Wogonin ( Fig. 1) is a flavone and has a variety of cardiovascular protective effect. It could regulate migration, proliferation and apoptosis of vascular smooth muscle cells. [6][7][8] Besides, wogonin could inhibit angiogenesis, suppress collagen deposition in cardiac fibroblasts and inhibit ischemic brain injury. [9][10][11][12] There is no evidence for vascular relaxation effect of wogonin. We describe here that wogonin, unlike oroxylin A, is an endothelium-and NO-independent vasodilatory flavonoid. One report demonstrated that wogonin offered a wide margin of safety.13) It has therapeutic potential for the treatment of cardiovascular and cerebrovascular diseases.

Summary Terminalia arjuna (T. arjuna) is an Indian medicinal plant credited with cardiotonic and cardioprotective properties; and baicalein is the active ingredient. To determine the possible mechanisms of protective action of T. arjuna and baicalein, we studied the reactions of T. arjuna extracts and baicalein with the biologically important superoxide (O2˙−) and singlet oxygen ( 1 O2) by measuring O2˙− or 1 O2 induced damage of lipids, lipid preoxidation, in the reaction mixture containing rat liver mitochondria and cardiac homogenate. Absorption of T. arjuna and baicalein in rat small intestine was also assessed. In addition, inhibitory effect of T. arjuna and baicalein on radiation-induced ROS generation in NIH3T3 cells and a cell-free chemical system, as well as on whole plasma oxidation were studied. Significant absorption of T. arjuna and baicalein from the intestine was observed. Baicalein is highly effective in inhibiting lipid peroxidation, reactive oxygen species generation, and plasma oxidation, even at low concentrations of 10-25 µM.
