Antiplatelet and anticoagulant activities of two phospholipase A2s purified from Cerastes cerastes venom: Structure‐function relationship

Abstract: This study aimed to elucidate anticoagulant/antiplatelet mechanisms of two previously purified PLA2s from Cerastes cerastes venom, here, termed Cc1‐PLA2 and Cc2‐PLA2. Both PLA2s present close molecular weights of 13,534 and 13,430 Da and Isoectric pH (pI) 7.38 and 7.86 respectively, for Cc1‐PLA2 and Cc2‐PLA2. These Ca2+‐dependent enzymes showed a high catalytic activity upon phospholipids, inducing indirect hemolysis, since they conserve the catalytic domain of PLA2s 26CYCGWGGKG34. They exhibited dual inhibiti… Show more

“…PLA 2 , here found only in the venom of N. fodiens , is widely distributed among elapid and viperid snake venoms and in scorpion venoms [ 27 , 53 , 54 ], and is also known for its cytotoxicity, including hemolytic activity [ 27 , 29 , 40 , 55 , 56 ]. For instance, PLA 2 from Cerastes ceratses venom produces indirect hemolysis [ 56 ]. Lazcano-Pérez et al [ 40 ] confirmed a dose-dependent hemolytic activity of cnidarian venom, which contains PLA 2 .…”

“…Cerastes is considered a venomous snake due to its hemotoxin venoms. In addition to SVMPs and hemotoxin, phospholipase A2 (PLA 2 ) was found to be the main constituent of the venom [ 53 ]. Here we reviewed four patients who suffered from Cerastes bites.…”

“…Whereas, unlike thrombin, these serine proteases are not able to activate FXIII required for stabilizing formed thrombus. In this case, the produced clots by SVTLEs are unstable and easily cleared [127,211]. These characteristics make SVTLEs good candidates as lead bio-compound therapeutics or diagnostics to dissolve undesirable embolus resulting in platelet hyperactivation and coagulation associated with COVID-19 pathogenesis.…”
