Antiplatelet Activity ofα.- andβ.-Amyrin, Isomeric Mixture fromProtium heptaphyllum.

Abstract: A mixture of triterpenes named a-and b-amyrin (AMI), isolated from the Brazilian medicinal herb Protium hetaphyllum (Aubl) March (Burseraceae), was evaluated for the ability to inhibit aggregation of human platelets induced by adenosine 5 0 -diphosphate (ADP, 1.5 and 3 mM), collagen, and arachidonic acid (AA) in vitro. The results showed that AMI significantly inhibited platelet aggregation (40, 64, and 60%) in the assay carried out with ADP (3 mM) as agonist, at the doses of 100, 150, and 200 mM, respectively… Show more

“…Alpha and beta amyrin: in vitro assays have shown that amyrins inhibited platelet aggregation induced by different agonists, 29 in particular β-amyrin inhibited collagen induced aggregation with IC50 lower than Aspirin. 30 α-Amyrin, has also shown to be a competitive inhibitor of PKC.…”

“…Vitor et al (2009) and Okoye et al (2014) reported that α-amyrin acetate ( 1 ) and α-amyrin ( 2 ) exhibited pronounced anti-inflammatory effects and suppressed the levels of inflammatory cytokines and COX-2 levels via inhibition of NF-κB activity and of signalling pathways involving phospho-cyclic AMP response element binding protein (Vitor et al 2009; Okoye et al 2014). Moreover, 2 inhibited platelet aggregation induced by adenosine 5′-diphosphate, collagen and arachidonic acid (Aragão et al 2007). Qi et al 2008 mentioned that stigmast-4,22-diene-3,6-dione ( 9 ) exhibited anti-larval activity.…”

“…Plants OSCs always possess promiscuous activities, and could cyclize 2,3-oxidosqualene to different conformations simultaneously. For example, amyrin synthase from Glycyrrhiza uralensis can produce not only α-amyrin but also β-amyrin [7]. Due to the multiple-activity of OSCs, more than 100 kinds of triterpenoid skeletons could be generated during cyclization.…”
