Antiplatelet activity of aporphine and phenanthrenoid alkaloids from Aromadendron elegans blume

Abstract: Six aporphine and one phenanthrenoid alkaloids isolated from Aromadendron elegans Blume were investigated for their ability to inhibit arachidonic acid (AA), collagen and ADP induced platelet aggregation in human whole blood. The antiplatelet activity of the compounds was measured in vitro by the Chrono Log whole blood aggregometer using an electrical impedance method. Of the compounds tested, (-)-N-acetylnornuciferine, (-)-N-acetylanonaine and 1-(N-acetyl-N-methylamino)ethyl-3,4,6-trimethoxy-7-hydroxyphenanth… Show more

“…The mean platelet aggregation in whole blood was measured as a change in impedance over 6 min after the addition of the inducers by comparison with that of a control group impedance. The final concentration of DMSO in the whole blood was 0.5% to eliminate the effect of the solvent on the aggregation (Jantan et al, 2006). Aspirin was used as a positive control.…”

“…The primary screening results reveal that oxoglaucine possesses strong anticancer activity, such as against HCT-8 (ED 50 = 1.00 µg/ml) and K B (ED 50 = 2.00 µg/ml) (Chang et al, 2002;Chen et al, 2002;Wu et al, 1989). In addition, oxoglaucine exhibits other important pharmacological activities including antiplatelet aggregation (Chang et al, 1998;Jantan et al, 2006), immunomodulatory activity (Ivanovska et al, 1997 and2000), treatment of adjuvant arthritis , antiinflammatory (Remichkova et al, 2009), antifungal activity (Clark et al, 1987). “…Various isoquinoline alkaloids are used as traditional medicines in the Orient because of their immunomodulatory, antiinflammatory and antibacterial effects (Brezová et al, 2004;Jantan et al, 2006). In addition, they decrease the activity of elastase reductions in which are known to be associated with inflammatory disease and the development of arthritis (Tanaka et al, 1993).…”
