SUMMARYTerrestrial plants have been demonstrated to be sources of antimalarial compounds. In Cuba, little is known about antimalarial potentials of plant species used as medicinals. For that reason, we evaluated the antimalarial activity of 14 plant species used in Cuba as antimalarial, antipyretic and/or antiparasitic. Hydroalcoholic extracts were prepared and tested in vitro for the antimalarial activity against Plasmodium falciparum Ghana strain and over human cell line MRC-5 to determine cytotoxicity. Parasite multiplication was determined microscopically by the direct count of Giemsa stained parasites. A colorimetric assay was used to quantify cytotoxicity. Nine extracts showed IC 50 values lower than 100 µg/mL against P. falciparum, four extracts were classified as marginally active (SI < 4), one as partially active (Parthenium hysterophorus) exhibiting SI equal to 6.2 and two extracts as active (Bambusa vulgaris and Punica granatum), showing SI > 10. B. vulgaris showed the most potent and specific antiplasmodial action (IC 50 = 4.7 µg/mL, SI = 28.9). Phytochemical characterization of active extracts confirmed the presence of triterpenoids in B. vulgaris and polar compounds with phenol free groups and fluorescent metabolites in both extracts as major phytocompounds, by thin layer chromatography. In conclusion, antimalarial use of B. vulgaris and P. hysterophorus was validated. B. vulgaris and P. granatum extracts were selected for follow-up because of their strong antimalarial activity.

Poincianella pluviosa is a tree whose stem bark is reputed to have antidiarrheal, wound healing, antitumoral and antimalarial properties. Considering that the leaves of the tree are a more abundant and renewable source than the stem bark, we chose to study the antimalarial potential of this organ part. The aqueous leaf extract exhibited a rich phenolic profile. It was active (IC 50 = 6.88 ± 1.64 µg mL -1 ) against chloroquine resistant Plasmodium falciparum (W2) without toxicity to HepG2 A16 cells. The organic fraction was more active (IC 50 = 1.83 µg mL -1 ) than the aqueous leaf extract itself. Ten phenolic compounds were obtained from the latter. This is the first report on the phenolic composition of P. pluviosa leaves. Ellagic acid (IC 50 = 0.215 ± 0.007 µg mL -1 ) proved to be 32 fold more active than the aqueous leaf extract itself. Considering their renewable aspect, leaves of P. pluviosa appear to be a more interesting source of antimalarial compounds than stem bark.Keywords: Poincianella pluviosa var. peltophoroides, leaves, ellagic acid, malaria, Plasmodium falciparum (W2) IntroductionA reduction in the incidence of tropical diseases such as malaria in poor countries is considered a major goal of the international community.1,2 In 2013, malaria was reported to be a threat to 198 million people causing 584,000 deaths in the world, mostly children under 5 years old in Africa. The impact of this disease is a major risk with the growth of world's population, mainly in regions without access to health services. The lack of sufficient financial resources and public health capacities in poor endemic countries led to the use of inefficient medicines and/or protocols resulting in an increase of mortality rate and spread of parasite resistance. 2,3 Malaria is an infectious disease caused by protozoa of the genus Plasmodium, which is transmitted by female Anopheles sp. mosquitoes. Five species of Plasmodium are able to infect humans, Plasmodium falciparum being considered the most dangerous. 4 Some clinical manifestations such as fever, chills, mild anemia and diarrheas are associated with malaria. 3,5 According to World Health Organization (WHO)-established protocols, 2 the treatment of uncomplicated Plasmodium falciparum malaria involves artemisininbased combination therapy with drugs such as mefloquine, piperaquine or the sulfadoxine/pyrimethamine combination. Dark berry fruits are one of the top 10 richest sources of dietary polyphenols and have been examined for their pharmacokinetic benefits in the human body related to absorption, digestion, metabolism, and excretion (ADME). With the expansion of the world wide web and rise of discretionary income in Europe and North America impacting the global food trade during the 21st century, several species of berries have become available for general consumption that may have previously been out of reach of the average consumer. Compared to their commercial counterparts, these berries contain many of the same polyphenols, and the possibility exists for the discovery of novel phenolic compounds that may affect the ADME process in a host-beneficial way.Several species have demonstrated antioxidant, antiobesogenic, antimicrobial, and anti-inflammatory properties through in vitro, animal studies, and human clinical trials. This review examines the available chemical compositions of several dark berries and their effect on the ADME process, their implication in host health effects, and the potential of these emerging species to suggest areas for future research.
