The main objectives of this work are to optimize the extraction parameters, to test the antioxidant activity of Aloe Vera extract and to study the impact of this extract on deteriorating molds of Algerian variety of wheat (CIRTA). The extraction was optimized by central composite design. Determination of the polyphenols, flavonoids, and proanthocyanidins was performed by using colorimetric assays. Identification and quantification of phenolic compounds were performed by RP-HPLC-UV method. The antioxidant activity was tested by three methods: 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,20-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and CUPRAC (Cupric reducing antioxidant capacity), the antifungal activity of Aloe Vera extract on isolated strains from durum wheat were tested by dilution in a solid medium method. The optimum of total phenolic got was1,044 x 104 µg GAE/g of dry extract. The extract is rich in polyphenols, flavonoids, and proanthocyanidins. The analysis of phenolic compounds of Aloe Vera by RP-HPLC-UV revealed seven phenolic compounds. Strong antioxidant activity was obtained for Aloe Vera extract. Purification and microscopic study of isolated strains gave the possibility of identifying four strains: Alternaria spp1, Alternaria spp2, Penicillium spp, and Aspergillus spp. the antifungal potential of Aloe Veravaries according to the fungal genera and the concentrations of extract used.

Humans are constantly faced with changing environmental conditions which often lead to the production of various free radicals which must be dealt with in order to ensure their survival. 1 Reactive oxygen species (ROS) such as superoxide radicals, hydroxyl radicals and peroxyl radicals, are natural byproducts of the normal metabolism of oxygen in living cells and they play important roles in cell signaling. These ROS also play an integral function in the pathogenesis of many acute and chronic human diseases including atherosclerosis, ageing, cancer, diabetes, immunosuppression and stroke. 2 The human body is equipped with an inherent antioxidative mechanism with which the deleterious effects of the ROS are waded off. An attempt has been made to evaluate antimicrobial, antioxidant and cytotoxicity properties of extracts from onion (Allium cepa L.), garlic (Allium sativum), leaves of guava (Psidium guajava), papaya (Carica papaya), tea (Camellia sinensis), baen (Avicennia alba) and keora (Sonneratia apetala), respectively to apply as natural preservatives for tomatoes. The air-dried plant materials of the respective plant species were subjected to ethanol-methanol extraction, concentrated and stored at 4 °C before use. The extracts were dissolved in 95% ethanol for analysis of antioxidant and antimicrobial properties. Of the extracts tested, tea extracts showed the highest zone of inhibition against several pathogenic bacteria (E. coli 35.0±3.2 mm; P. aeruginosa 29.3±2.6 mm; S. typhi 28.4±2.1 mm and S. pyogenes 27.7±3.7 mm) using the disc diffusion method. In regard to DPPH free radical scavenging assay, keora and guava extracts showed the highest percentage of radical scavenging activity with the values of 89.64± 0.18 and 89.39± 0.88, respectively, which were in agreement with higher total antioxidant capacity (TAC) of these extracts obtained by the phosphomolybdenum method. Brine shrimp lethality bioassay for cytotoxicity assessment showed LC50 of 132.54 ± 18.99 µg/mL for the leaf extract of keora which was found to be most toxic among all studied extracts. The initial results indicated that the extracts could be used for food preservative applications based on the antimicrobial, antioxidant and cytotoxicity properties of the tested extracts. However, efficacy, stability and safety issues need to be addressed with both in vitro and in vivo studies.
