We describe the characterization of the centesimal composition, mineral and phenolic content of Eugenia uniflora fruit and the determination of the antioxidant, antimicrobial and quorum quenching activities of the pulp phenolic extract. Centesimal composition was determined according to standard methods; trace elements were measured by total reflection X-ray fluorescence spectroscopy. The phenolic compounds were extracted by solid-phase chromatography and quantified by spectrophotometry. Antioxidant activity was determined by using 3 different methods. Antimicrobial activity was evaluated against a panel of foodborne microorganisms and antiquorum sensing activity in Chromobacterium violaceum was performed by measuring inhibition of quorum sensing dependent violacein production. The centesimal composition (per 100 g of pulp) was as follows: protein 3.68 ± 0.21 g, lipids 0.02 ± 0.03 g, carbohydrates 10.31 g and fiber 2.06 g. Trace elements (mg/g of pulp) were determined as: K 0.90, Ca 3.36, Fe 0.60, Zn 0.17, Cl 0.56, Cr 0.06, Ni 0.04, and Cu 0.07. The pulp is a source of phenolic compounds and presents antioxidant activity similar to other berries. The fruit phenolic extract inhibited all tested bacteria. We also found that the fruit phenolic extract at low subinhibitory concentrations inhibited up to 96% of violacein production in C. violaceum, likely due to the fruit's phenolic content. This study shows the contribution of E. uniflora phenolic compounds to the antioxidant, antimicrobial and the newly discovered quorum quenching activity, all of which could be used by the food and pharmaceutical industries to develop new functional products.

The aim of the present study was to investigate the biological activities of the ultrasound‐assisted extracts obtained from pulp and seed of jujube (Ziziphus jujuba) fruits. To reach this purpose, total phenolic content (TPC), total flavonoid content (TFC), total pro‐anthocyanin, DPPH radical scavenging activity, rancimat test, as well as antimicrobial activity and cytotoxicity test of both jujube pulp and seed extracts were evaluated. Total phenolic content (TPC), total flavonoid content (TFC), and total pro‐anthocyanin in pulp extract were higher than those obtained from seed extract. In addition, DPPH radical scavenging activity of pulp extract (IC50 = 53.97 µg/ml) was higher than that of seed extract (IC50 = 88.68 µg/ml). Furthermore, the highest antimicrobial activity was observed against Escherichia coli and Staphylococcus aureus (MIC = 20 mg/ml) for both seed and pulp extracts. In vitro cytotoxicity evaluation on seven cell lines revealed that pulp and seed extracts of jujube had no cytotoxic activity. The present results suggested the promising antioxidant properties of jujube, which can be used in the fabrication of functional bioactive ingredients for different purposes.

Effects of different extraction methods on the phenolic profile, antioxidant and antimicrobial activity of the coffee grounds and coffee silverskin (Coffea arabica L.)

Background During coffee processing, a large volume of waste is generated. This study investigated the phenolic profile, antioxidant and antibacterial activity of coffee grounds and silverskin using different extraction methods. Results Ultrasound‐assisted extraction (UAE), enzyme‐assisted extraction (EAE), and ultrasound‐assisted enzyme extraction (UAEE) were effective for extraction of polyphenols, with UAE being the method that promoted the highest extraction of polyphenols. The coffee ground and coffee silverskin extracts showed high antioxidant capacity for the three methods ABTS, DPPH, and ORAC (4013.36–3241.89 μmol Trolox/g; 1.82–2.06 IC50 μg/mL; and 26.60–16.20 μmol Trolox/mL, respectively). The phenolic profile demonstrated that most of the compounds in both matrices were chlorogenic acid, caffeic acid, 2,4‐DHBA, 2,5‐DHBA, and 3,4‐DHB. EAE promoted a greater release of polyphenols in the coffee ground (2676.42 μg/g), while UAEE was more effective for the coffee silverskin (2111.54 μg/g). The Principal Component Analysis grouped the samples according to the extraction method, based on their content and phenolic potential, for coffee ground and coffee silverskin. As for antibacterial activity, the most expressive result was for Gram‐positive bacteria. For both matrices, when extracted by the UAEE method, they presented the minimum inhibitory concentration to inhibit the growth of Bacillus subtilis, Bacillus cereus, and Listeria monocytogenes. Conclusion The extraction methods used are environmentally safe alternatives, enabling the reuse of natural compounds and contributing to the circular economy, seeking to transform waste into raw material for the extraction of compounds of interest and possible subsequent application in agriculture as biofertilizers or bioherbicides.
