Antioxidant and Anti-Proliferative Activity of Essential Oil and Main Components from Leaves of Aloysia polystachya Harvested in Central Chile

Abstract: The aim of this study was to determine, first, the chemical composition of Aloysia polystachya (Griseb) Moldenke essential oil, from leaves harvested in central Chile; and second, its antioxidant and cytotoxic activity. Eight compounds were identified via gas chromatography–mass spectrometry (GC–MS) analyses, with the most representative being R-carvone (91.03%), R-limonene (4.10%), and dihydrocarvone (1.07%). For Aloysia polystachya essential oil, antioxidant assays (2,2-diphenyl-1-picrylhydrazyl (DPPH), H2O2… Show more

“…A standard curve was prepared with Trolox (50-800 μ M). The results were expressed as μ mol equivalents of Trolox per gram of essential oil ( μ mol TE/g EO) [ 28 ]. To calculate the half inhibitory concentration (IC 50 ), the linear regression method was used based on the different concentrations of the EO.…”

“…However, the individual compounds (carvone and dihydrocarvone) did not exhibit cytotoxic activity against the tumor lines tested. 63 The anti-tumor activity of monoterpenes seems to be related to different mechanisms, including induction of enzymes from phases I and II of carcinogenic metabolism, cell cycle arrest, apoptosis induction, ROS generation, inhibition of post-translational isoprenylation of cell-growth regulation proteins, and modulation of multiple molecular targets. 90,91 Vinothkumar et al uncovered the anticancer effects of carvone against 1,2-dimethylhydrzine-induced rat colon carcinogenesis through preventing pre-neoplastic lesions, oxidative stress and abnormal stimulation of biotransforming enzymes.…”

“…The Aloysia polystachya essential oil with 91.03% carvone showed good antioxidant activity by different antioxidant assays, including 2,2-diphenyl-1-picrylhydrazyl (DPPH), H 2 O 2 , ferric reducing antioxidant power (FRAP), and total reactive antioxidant potential (TRAP) assay. 63 Sabir et al described that carvone isolated from Zanthoxylum alatum exhibited antioxidant and free radical scavenging activities. The isolated carvone could inhibit Fe( ii ) and sodium nitroprusside (SNP) induced thiobarbituric acid reactive species (TBARS) in rat brain, as well as cause the scavenging of DPPH radicals and the reduction of molybdenum.…”

“…The fresh leaves (500 g) of L. sempervirens were submitted to hydrodistillation for 4 h. The distilled leaves were extracted using ethyl acetate, dried over anhydrous sodium sulfate, and the solvent evaporated. The EO was analyzed and its components identified by gas chromatography/mass spectrometry (GC/MS) using previously standardized instrumentation and methodology [32].…”
