Purpose: Reactive oxygen species and the free radicals are implicated in many medical disorders as well as in the process of aging. Due to the toxicity of synthetic drugs there is much interest in natural products that could combat diseases. Furthermore side effects and resistance to antibiotics are well documented and as such there is much interest in herbal remedies that have antimicrobial activity. The objective of this research is to investigate the antioxidant and antibacterial potential of the extracts of the seed and peel of Spondias dulcis (Amberalla) fruit. Method: The percentage radical scavenging activity was determined using 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging assay. The reducing potential of the extracts were determined using Ferric ion reducing assay. The total phenol and total flavonoid contents were obtained from calibrations plots of Gallic acid and Quercetine respectively. The antibacterial assays against Staphylococcus aureus, Escherichia coli and Bacillus subtilis. were carried out using Agar well method. Results: The antioxidant activity, total flavonoid and phenolic contents of the ethyl acetate, hexane and dichloromethane extracts of the Sri Lankan variety of Spondias dulcis was investigated. Of all extracts the highest activity was shown by the peel extracts of the S.dulcis fruit. The dichloromethane extract of the peel of the fruit showed the maximum activity in DPPH radical scavenging assay (93.40±0.02%) followed by the hexane extract of the peel (91.10±0.01%) compared to butylated hydroxy toluene (BHT) (74.26±0.01%) at the same concentration(127.5 µg ml -1 ). In the Fe 3+ reducing power assay the dichloromethane and ethyl acetate extracts of the peel gave values of 0.35±0.12% and 0.30±0.05% respectively compared to BHT (0.71±0.04%) at the same concentration(3.33 µg ml -1 ).The highest phenolic content was found in the ethyl acetate extracts of peel (463.68±0.02 mg (GAE/g). The maximum flavonoid content was seen in the hexane extract of the peel (1133.77±0.01mg (QE/g). However, the antibacterial assay of the peel and the seed extracts did not show a significant activity against the bacterial species; Staphylococcus aureus, Escherichia coli and Bacillus subtilis. Conclusion: Our results show that the non-utilized parts of the S. dulcis fruit contains high amount of antioxidant properties and may have the ability to reduce the risk of oxidative stress and defense against degenerative diseases.

The genus Jeffreycia consists of about 109 species to have medicinal properties. Jeffreycia zeylanica (Family: Asteraceae) is commonly referred to as “Heen-botiya, Papula or Pupula in Sinhala and Kappilay or Kuppilay in Tamil”. It is an endemic herbaceous plant in Sri Lanka. Synonyms of Jeffreycia zeylanica are Cacalia zeylanica Kuntze, Eupatorium zeylanicum L., Gymnanthemum zeylanicum (L.), Vernonia zeylanica (L.). This plant can create secondary metabolites that may have substantial bioactivity. Extracts from leaves, stems and aerial parts are rich in alkaloids, flavonoids, steroids, triterpenoids, polyphenols, and saponins. These bioactive compounds lead to various pharmacological properties of Jeffreycia zeylanica. It is utilized in both Ayurvedic and folk medicine in a variety of methods to treat various illnesses, such as bone fractures, eczema, asthma, diarrhea, wound, and abscesses also as an anti-venom. Several studies have been carried out on this plant, including the evaluation of anti-inflammatory activity, antinociceptive activity, antioxidant activity, and anti-tumor activity. The lack of considerable toxicity associated with the plant Jeffreycia zeylanica makes it a choice for future development of therapeutic products, with the combination of traditional and novel technologies. A significant number of presently used antioxidant, anti-inflammatory, anthelminthic, and antitumor agents are compounds isolated from plants or their synthetic or semisynthetic derivatives.

This study was undertaken to analyse the potential bioactivities including antibacterial, antioxidant and antidiabetic derived from the methanolic extract and the column chromatography ethyl acetate fraction (AcOEt Fr) of Horsfieldia spicata leaves. Methanolic extract and 4 other fractions was calculated for total phenol and flavonoid contents along with tested for antibacterial, antioxidant and antidiabetic properties. Interestingly, the AcOEt Fr had the highest value for total flavonoid content and the best antioxidant, and antidiabetic activities. Therefore, the AcOEt Fr was further separated using column chromatography technique for obtaining 9 selected fractions namely fraction 1 (F1) ‐ fraction 9 (F9) which were further tested. The results showed that the AcOEt column chromatography fractions namely F2, F3, F4 and F6 had the best clear inhibition antibacterial value against all bacterial tested. In addition, these fractions also exhibited better Minimum Inhibitory Concentrations (MIC) and Minimum Bactericidal Concentrations (MBC) values than others. Antioxidant, 2,2‐diphenylpicrylhydrazyl (DPPH) assayed indicated that AcOEt Fr had the strongest IC50 value of 47.30 μg/mL. Further, F4 column chromatography fraction showed the best inhibition against α‐Glucosidase enzyme related to antidiabetic activity with an IC50 value of 6.11 μg/mL. Liquid chromatography tandem‐mass spectrometry (LC/MS/MS) analysis identified that F4 derived from AcOEt fraction had several compounds belonging to the flavonoid and phenolics such as 3′,5‐dihydroxy‐7,4′‐dimethoxyflavone, 5,7‐dihydroxy‐3‐(4′‐hydroxybenzyl)chromone, and Kadsurenin I.
