Antioxidant activity and cytotoxicity of the edible mushroom, Pleurotus cystidiosus against Hep-2 carcinoma cells

Abstract: This study was undertaken to evaluate the antioxidant activity and cytotoxicity of Pleurotus cystidiosus, an edible mushroom, against Hep-2 cancer cells. Fresh P. cystidiosus mushroom was extracted with acetone (fraction A). Fraction A was extracted into hexane, dichloromethane and ethyl acetate successively and the remaining fraction was labeled as "A4". Fraction A4 was further separated into three fractions, A4-1, A4-2 and A4-3 using a reverse phase column. 1,1-diphenyl-2picrylhydrazyl (DPPH) radical scaveng… Show more

“…The past decade has witnessed the overwhelming interest in isolation and characterization of bioactive compounds from medicinal mushrooms due to the fact that natural products of mushroom origin are found to possess a remarkably high pharmacological index [26, 27]. The mounting evidences from various scientific studies regarding the applications of mushroom extracts and substances in the development of anti-cancer therapeutics unarguably make it a fast-track research area worth mass attention.…”

“…Pleurotus cystidiosus is another important edible mushroom which possesses antiproliferative activity. It has been shown cytotoxic properties against Hep-2 cells with an EC 50 value of 3.6 mg/mL for a fraction of methanol:water (7:3) [ 33 ]. MEFCA also exhibits strong antiproliferative activity compared to the activity shown by P. cystidiosus.…”

“…Human cancer cell line, HEp 2 (epidermoid carcinoma, larynx, adherent, monolayer) was cultured in vitro in a growth medium (GM) consisting of 10% fetal bovine serum (FBS) in Dulbecco's modified Eagle's medium (DMEM) supplemented with L-glutamine, antibiotics (50 IU/ mL penicillin and 50 μg/mL streptomycin) and 7.5% sodium bicarbonate in plastic tissue culture flasks, and cultures were incubated in a humidified incubator (Heal Force-HF151) maintained at 37°C with 5% CO 2 [10]. Cells were transferred into 96 well plates, with clear bottoms, and incubated overnight at 37°C with 5% CO 2 in a humidified incubator.…”
