Antinuclear antibodies in the relatives of patients with systemic sclerosis

Abstract: Antinuclear antibodies were studied by indirect immunofluorescence and double immunodiffusion in 21 patients with systemic sclerosis and 35 of their relatives. When HEp-2 cells were used as the substrate, the frequency of antinuclear antibodies in the patients' sera was 100% and that in the relatives was 26%. When rat liver sections were used, the values were 86% and 17%, respectively. Anticentromere antibody was detected in the serum from the mother of one patient whose serum had anti-Scl-70 antibody. Antibod… Show more

“…However, wc and other investiga tors have discovered that this antibody can also be found in patients suffering from other diseases. The antibody has frequently been detected in cases of primary biliary cirrhosis [ 10. 111 and in cases of Raynaud's phenomenon alone [2,7,12]. Additionally, this antibody has been reported to be present occasionally in cases of systemic lupus erythematosus [2,7,13], localized scleroderma [14][15][16] and the blood relatives of patients with systemic scle rosis [5.…”

“…Indirect immunofluorescence studies were performed on IIEp-2 cells, as described previously [5][6][7], HEp-2 cells grown directly on glass slides were purchased from Anti bodies Inc. (Davis, Calif.. USA). The fluorcsccin-isothiocyanatcconjugated goat antihuman immunoglobulin antiserum used came from Eiken Chemical (Tokyo, Japan).…”

“…The presence of anti-RNP antibody alone strongly suggests a diagnosis of MCTD. Scleroderma (Scl-70) autoantibodies are a specific marker for scleroderma and are found in up to 60% of patients diagnosed with this disorder (2,4,27,29,30).…”

“…ELISA test is reported to give false positive results in sera with antinuclear antibodies (ANA) or rheumatoid factor (RF) (Walls et al 1977). However, inspite of lack of accurate data on the incidence of RF and ANA in Japan, antinuclear antibodies in the population are very rare (Takehara et al 1985). Moreover, it has been reported that although the RF may affect the degree of positiviy detected for a given sample it does not affect positiviy itself (Woodward 1982).…”
