Antimicrobial Resistance and Type III Secretion System Virulotypes of Pseudomonas aeruginosa Isolates from Dogs and Cats in Primary Veterinary Hospitals in Japan: Identification of the International High-Risk Clone Sequence Type 235

Abstract: Pseudomonas aeruginosa is an environmentally ubiquitous and important opportunistic human pathogen responsible for life-threatening health care-associated infections. Because of its extensive repertoire of virulence determinants and intrinsic and acquired resistance mechanisms, the organism could be one of the most clinically and epidemiologically important causes of morbidity and mortality.

“…An earlier report from a tertiary hospital in Daejeon, Korea indicates a lack of any resistance genes in ST1455 but on the contrary, all our ST1455 isolates were carbapenem-resistant [41]. Carbapenem resistance has been observed in ST3045 isolates isolated from dogs in Japan and it was true for our isolates except that they were isolated from the human host [42]. P. aeruginosa strain ST663 has been observed in France and also been isolated from healthy human volunteers in Spain where it has an imipenem breakpoint of 4mg/L which groups it into sensitive category.…”

“…However, some rare cases of carbapenemase expression, especially VIM-2, have been reported in different countries in P. aeruginosa strains isolated from dogs, cattle, and fowl [ 90 , 91 ], but to our knowledge not yet in horses. Some studies even suggest the existence of zoonotic transmission from animals to humans [ 92 ] and from humans to animals [ 93 , 94 ], but in an anecdotal manner, including the case of a transmission of a VIM-2 strain from humans to animals in Brazil [ 95 ].…”

“…P. aeruginosa expresses six types of secretion systems ( Figure 1 ) that participate in the transport of several virulence factors, including enzymes, exotoxins, and other proteins [ 20 , 44 , 48 , 54 , 55 , 56 , 57 , 58 ].…”

“…In two-step secretion systems, the protein is temporarily stored in the periplasmic space located between the inner and outer membranes, before being released to the external environment ( Figure 1 ). These differences affect the efficiency and specificity of protein transport [ 20 , 44 , 55 , 56 , 57 , 58 ].…”
