Antimicrobial and Anticoagulant Activities of N -Chlorotaurine, N , N -Dichloro-2,2-Dimethyltaurine, and N -Monochloro-2,2-Dimethyltaurine in Human Blood

Abstract: The aim of this study was to determine the potential application of N-chlorotaurine (NCT), N,N-dichloro-2,2-dimethyltaurine (NVC-422), and N-monochloro-2,2-dimethyltaurine (NVC-612) as catheter lock solutions for the prevention of catheter blockage and catheter-related bloodstream infections by testing their anticoagulant and broad-spectrum antimicrobial activities in human blood. NCT, NVC-422, NVC-612, and control compounds were serially diluted in fresh human blood to evaluate the effects on prothrombin time… Show more

“…NCT was investigated for nearly 15 years, and its anti-infective usefulness has been confirmed to date by numerous studies (reviewed in reference 49). DM-NCT and DM-NDCT, on the other hand, are new compounds, whose anti-infective properties are currently explored in detail, yielding promising results, which confirm them as equivalent to NCT (23,25,(50)(51)(52). Specifically, DM-NDCT (NVC-422) is active against a broad range of Gram-positive and Gram-negative bacteria, including drug-resistant pathogens (25), without generating resistance in multiplepassage studies (53).…”

“…An impressive example concerns NCT and the very reactive hypochlorous acid (HOCl). While the consumption of HOCl is twice as high (45), the bactericidal effect, however, is approximately 10,000-fold higher (51,67,68). This astonishing result can be reduced to both the faster penetration of HOCl and its superior oxidizing activity (see above), which causes immediate inactivation of S-H and S-CH 3 functions of vitally essential enzymes.…”

“…Its bactericidal activity against S. aureus and S. epidermidis in biofilm (reduction of CFU to the detection limit by 1% NCT needed about 30 to 60 min [ Fig. 3]) appears approximately 3 to 6 times slower than that against planktonic bacteria (10 min for a reduction to the detection limit [9,10]) but is still sufficient. Remarkably, viable counts of all tested bacteria in biofilm were reduced by at least 1 power of 10 after 15 min of incubation in 1% NCT, so that an impact on biofilm by irrigations with NCT can be conceived in vivo.…”

“…The plates were incubated at 37 ± 1 °C for 24 h and the numbers of fungal CFU were counted and assessed, considering the duplicity and the dilution [6]. …”
