Antimicrobial Activities and Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry of Bacillus Isolates from the Marine Sponge Aplysina aerophoba

Abstract: The aim of this study was to isolate bacteria that are resistant to the strong antimicrobial metabolites characteristic of Aplysina aerophoba. For this purpose, bacterial isolation was performed on agar plates to which sponge tissue extract had been added. Following screening for antifungal and antimicrobial activities, 5 strains were chosen for more detailed analyses. 16S ribosomal DNA sequencing revealed that all isolates belonged to the genus Bacillus, specifically B. subtilis and B. pumilus. Using a combin… Show more

“…The production of bioactive compounds by Bacillus species is well established (Korzybski et al, 1978;Naruse et al, 1990;Munimbazi & Bullerman, 1998). These classes of bioactive compounds act as antifungal peptides, antifungal lipopeptides and antimicrobial polypeptides (Pabel et al, 2003). To characterize antimicrobial activity of B. pumilus MAIIIM4a, the isolate was tested against three phytopathogenic fungi: R. solani, P. aphanidermatum and S. rolfsii.…”

“…ICMS spectra in the range below 2 kDa identify characteristic metabolites, e.g. of bacilli (Williams et al, 2002;Pabel et al, 2003) and cyanobacteria (Erhard et al,1997;Welker et al, 2002Welker et al, , 2004a, and we show here that highly characteristic peptaibol spectra can readily be obtained from microgram amounts of mycelia, thus providing characteristic metabolite fingerprints. A comparison of these fingerprints with molecular phylogenies of the species documents some correlation between types of peptaibol formed and phylogeny.…”
