Antimalarial Pyrido[1,2-a]benzimidazoles

Abstract: A novel class of antimalarial pyrido[1,2-a]benzimidazoles were synthesized and evaluated for antiplasmodial activity and cytotoxicity following hits identified from screening commercially available compound collections. The most active of these, TDR86919 (4c), showed improved in vitro activity vs the drugresistant K1 strain of Plasmodium falciparum relative to chloroquine (IC 50 = 0.047 μM v 0.17 μM); potency was retained against a range of drug-sensitive and drug-resistant strains, with negligible cytotoxicit… Show more

“…A multifactorial optimisation campaign, identified compound 61 as a lead, which possessed excellent, albeit slightly varied in vitro anti‐plasmodial activity against several drug‐sensitive and ‐resistant strains, with negligible mammalian cytotoxicity. However, while this activity translated into good in vivo efficacy using several different salts of 61 , there were concerns relating to pharmacokinetic parameters . A subsequent follow‐up, sought to develop analogues with improved pharmacokinetic properties, as well as exploring additional features of the SAR of this class .…”

“…The benzimidazole moiety is amongst the diversely active scaffolds as antifungal, antibacterial, antiviral, anticancer and antimalarial agents. [1][2][3][4] Therefore, developing new approaches towards the synthesis of benzimdazole fused heterocycles have always been on need. 5 Also, indeno and isoquinolino fused heterocycles have gained greater attention because of their medicinal and synthetic importance over years and the broad range of biological activity.…”

“…Benzimidazole based compounds have been reported to exhibit FP‐2 inhibitory activity. On the basis of the core pharmacophoric features of these benzimidazole derivatives, novel benzimidazole acrylonitriles derivatives have been designed and synthesized. In this paper, we report synthesis of the designed compounds as inhibitors of falcipain‐2 enzyme and hemozoin formation which could be developed as potential antimalarial drugs.…”
