Antimalarial and β-hematin formation inhibitory activities of chromone derivatives

Abstract: A series of chromone compounds were evaluated as new potential antimalarial agents using in vitro antimalarial activity assay. The most potent compound 36 with an IC 50 of 0.95 µM was shown to be more potent than primaquine and tafenoquine (IC 50 2.41 and 1.95 µM, respectively). β-Hematin formation inhibitory activity test and stoichiometry determination have also been performed to investigate the preliminary mechanism of antimalarial activity of the studied compounds.

“…Chromone containing natural and synthetic molecules exhibited important biological activities [1] . A wide range of medicinally significant properties displayed by chromone scaffolds are neuroprotective, [2] anti‐malarial, [3] anti‐fungal, [4] anti‐bacterial, [5] and anit‐HIV [6] . Chromone derivatives have also shown anticancer and antiviral potential [7–11] .…”

“…Chemically, chromone is 4Hchromen-4-one, also known as 4H-1-benzopyran-4-one which is a isomer of coumarin and a core part of various flavonoids. [131][132][133] Chromones exhibit wide range of biological applications like anti cancer, [134] anti bacterial, [135] anti fungal, [136] anti oxidant, [137] anti inflammatory, [138] anti-HIV, [139] anti malarial, [140] neuroprotective effects. [141] Chromones are structurally similar to chalcones and coumarins and having selective inhibition towards MAOÀ B.…”
