Antihypertensive Effect of 5-HT1A Agonist Buspirone and 5-HT2B Antagonists in Experimentally Induced Hypertension in Rats

Abstract: We investigated the antihypertensive effect of 5-HT1A agonist (buspirone) and 5-HT2B antagonists (SB204741 and SB200646) in deoxycorticosterone acetate (DOCA)-salt-induced hypertensive rats. Experiments were divided into two sets: in the first set, sham-operated control and DOCA-treated hypertensive rats received buspirone (1 mg/kg/day p.o. for 4 weeks) and in the second set, in vivo and in vitro studies were carried out. In the case of in vivo studies, sham-operated control and DOCA-trea… Show more

“…That led to decrease in SBP[45] and improve diastolic function in the left ventricle, which led to decrease in DBP. [46] Bu as reported by Shingala and Balaraman, 2005,[4] has an antihypertensive effect through aortic effect and vascular activity (DBP) Furthermore, it was reported that Bu reduced the SBP. [47]…”

“…However, conflicting reports by Hanson et al [30], Taylor et al [31], and Unrug et al [32] suggest a decrease in mean arterial blood pressure and HR. Further, it has also been reported that chronic administration of buspirone in DOCA induced hypertensive rats significantly reduces BP and 5-HT1A agonists might possess antihypertensive action [33]. The effect of buspirone on CVS functions seems to be dependent on species, route, and dose [30].…”

“…The same procedure was followed in rats, except the tightening and the releasing of the cannula were considered as Sham operated animals. The carotid artery was cannulated using transducer (SL 13L, Biopac Systems) for measuring the hemodynamic parameters 10 . Parameters like systolic (SBP), diastolic (DBP) and mean arterial blood pressure (MAP) were observed.…”
