Antigen-specific T-cell factor in cell cooperation. Mapping within the I region of the H-2 complex and ability to cooperate across allogeneic barriers.

Abstract: Further mapping of the mouse T-cell factor specific for poly(Try,Glu)-polyD-LAla--polyLys is reported. It is shown to be a product of the I-A subregion of the H-2 complex by the use of antisera either raised specifically against or made specific, by absorption, for different regions of the H-2 complex. The factor cooperates across allogeneic barriers, e.g., when factor produced by one strian is combined with bone marrow cells of other H-2 incompatible strains.

“…It is tempting to generalize these findings and to consider that the I-J subregion is specialized to code for suppressive molecules. On the other hand, the specific helper factors described by Taussig et al (16) and Isac et al (17), appear to be coded by the I-A subregion, stressing again the specialization of different I subregions of the H-2 complex and the molecular distinction between helper and suppressor materials. Different I subregions therefore appear to be specialized in coding for the specific helper or suppressor factors recently described.…”

“…cluding helper T-cell-replacing factors (17,18) and suppressor factors (19,20). These molecules share certain properties: mol wt of approximately 50,000, antigen-binding sites, the lack of Ig determinants, and the presence of determinants recognized by alloantisera specific for I-region gene products.…”

“…Whether the supernate of alloantigen-activated cells contains Ia moieties is not presently known. However, complex molecules containing such/-region products have been identified in other T-cell factors which mediate regulatory cell interactions in antibody synthesis (12)(13)(14). Although MLR responder cells have been reported to lack Ia antigen (7), it is possible that such molecules are exposed or altered to a definable configuration subsequent to antigen stimulation, providing the appropriate site of interaction with an Ia-containing suppressor factor.…”
